蛙皮素降温作用与AC、cAMP、PKA、NF-κB关系的实验研究

【摘要】 目的该项研究旨在观察蛙皮素的降温作用与EP3发热信号转导通路之间的关系。方法通过侧脑室插管给予IL-1β制备发热大鼠模型,采用Elisa、免疫组化染色、Western blot和Real time-PCR方法检测下丘脑组织中腺苷酸环化酶(adenylate cyclase,AC)、环磷酸腺苷(cyclic adenosine monophosphate,cAMP)、蛋白激酶A(protein kinase A,PKA)、核转录因子-κB(NF-κB)的表达情况。结果 a.正常组和蛙皮素组大鼠下丘脑组织中AC、cAMP、PKA、NF-κB的表达量明显低于发热组。b.在总蛋白、胞质蛋白、胞核蛋白水平上,发热组大鼠明显高于正常组和蛙皮素组;正常组、蛙皮素组NF-κB的mRNA的表达量明显低于发热组。结论蛙皮素的降温作用与AC、cAMP、PKA和NF-κB的表达量变化有关,可能是通过对EP3信号转导通路的影响而实现的。

【Abstract】 Objective To investigate the relationship between the hypothermic effect of bombesin and the EP3 signal transduction pathway.Methods The rat model of fever was prepared by IL-1β in the lateral ventricle cannula.The expressions of AC,cAMP,PKA and NF-κB in hypothalamus were detected by Elisa,Immunohistochemical staining,Western blot and Real time-PCR.Results The expression levels of adenylate cyclase,cyclic adenosine monophosphate,protein kinase A and NF-κB in the hypothalamus of the normal group and the bombesin group were significantly lower than those of the fever group.The levels of total protein,cytoplasmic protein and nucleoprotein in the rats in the fever group were significantly higher than those in the normal group and the bombesin group.The expression of NF-κB mRNA in the normal group and the bombesin group was significantly lower than that in the fever group.Conclusion The hypothermic effect of bombesin is related to the changes of AC,cAMP,PKA and NF-κB expression,which may be achieved through the influence of EP3 signal transduction pathway.
