背景荧光猝灭-免疫层析法检测牛奶中卡那霉素的方法建立

【摘要】 目的建立基于背景荧光猝灭-免疫层析技术快速定量检测牛奶中卡那霉素的方法。方法根据卡那霉素抗原和抗体的活性,筛选出卡那霉素抗原和抗体在检测系统中最佳的比例浓度,按照筛选出的最佳检测系统比例进行牛奶样品中卡那霉素浓度测定及完成方法学验证。结果以最佳方法测定牛奶中卡那霉素的浓度,其测定范围为0.231 9~60.01μg·L-1,最低检测限为0.200 3μg·L-1;牛奶样品中高、中、低卡那霉素测定的平均加标回收率分别为98.9%、102.3%、95.9%,RSD为2.36%、4.89%、1.41%。牛奶样品采用本法与国标方法的检测结果无显著性差异(P>0.05)。结论本方法可以确定为一种的检测牛奶中卡那霉素的专属性强、灵敏度高、简便、快速、准确的的新方法。

【Abstract】 Objective Aquantitative determination card of Kanamyci(KaA) was developed by utilizing the background fluorescence quenching immunochromatographic assay(b FQICA)in order to detect the content of KaA in pork rapidly.Methods Based on the activities of KaA antigen and antibody,the best concentration of antigen of KaA andantibody of KaA in the detection system were screened.Methodology validation of KaA in milk and sample determination was conducted by this detection system.Results The concentration of KaA in this detection system ranged from 0.2319~60.01 μg·L-1.The recoveries at three spiked levels ranged 98.9%、102.3%、95.9%,with RSD of 2.36%、4.89%、1.41%.The result of t-test showed that there was no significant difference between measured values of KaA in sample by bFQICA and by National Standards of the People′s Republic of China(P>0.05).Conclusion This method is accurate,sensitive,rapid and precise for detecting KaA in milk.
