支气管肺炎大鼠的肺炎影像学与高尔基体应激水平和肺功能的关系

【摘要】 目的探究支气管肺炎大鼠的肺炎影像学与高尔基体应激水平和肺功能的关系。方法 24只10~12周龄雌性Wistar大鼠用于本次的实验,实验前采用脂多糖(LPS)鼻内滴注诱导支气管肺炎模型,造模成功后作为模型组,另取正常大鼠鼻内滴注0.9%氯化钠溶液为对照组,每组各12只。正电子发射断层扫描(PET)扫描仪进行肺炎影像学评估。组织病理学对肺组织炎症损伤进行评分。使用酶标仪检测大鼠的髓过氧化物酶(MPO)活性。实时定量聚合酶链反应(RT-PCR)实时分析,白细胞介素-1(IL-1)、IL-6、肿瘤坏死因子-α(TNF-α)的mRNA表达。使用恒定相位模型评估肺力学评估。结果注射FEDAC后的6、12、24 h后模型组较对照组大鼠肺部放射性吸收量显著增加(P <0.05)。与对照组大鼠相比,模型组刺激了肺泡充血,并且主要增加了肺泡壁的厚度以及炎性细胞的浸润或聚集。模型组较对照组炎症综合评分[(6.84±1.37) vs.(1.37±0.26)分)]、活性氧(ROS)含量[(4.19±1.24 vs.(1.57±0.23)nmol/L)]、髓过氧化物酶(MPO)活性[(6.31±1.27) vs.(1.08±0.14) nmol/L)]、炎症指标IL-1(1.86±0.24 vs.0.94±0.08)、IL-6(1.95±0.26 vs.1.05±0.13)、TNF-α(2.01±0.35 vs.1.14±0.14)的mRNA表达以及肺功能指标肺阻力(Rrs)[(2.15±0.14) vs.(1.24±0.05) cmH2O.s/mL)]、肺弹性(Ers)[(41.67±5.08) vs.(41.67±5.08) cm H2O.s/mL)]、牛顿阻力(Rn)[(1.74±0.12) vs.(0.78±0.08) cm H2O.s/mL]和组织阻尼(Gtis)[(8.36±1.29) vs.(4.29±0.67) cm H2O.s/mL)]均显著升高,差异有统计学意义(P <0.05)。结论支气管肺炎大鼠高尔基体应激水平改变,促炎细胞因子的表达增加,肺泡壁的厚度增加以及炎性细胞的浸润或聚集,从而导致肺功能降低,PET是用于监测肺部炎症进展的有用工具。

【Abstract】 Objective To explore the relationship between pneumonia imaging and Golgi apparatus stress level and lung function in bronchopneumonia rats.Methods Twenty four female Wistar rats aged 10 to 12 weeks were used in this experiment.Before the experiment,intranasal infusion of lipopolysaccharide(LPS) was used to induce the bronchopneumonia model.After the model was successfully established,it was used as the model group,and normal rats were injected with normal saline as the control group.Positron emission tomography(PET) scanner for imaging evaluation of pneumonia.Histopathology scores the inflammatory damage of lung tissue.A microplate reader was used to detect the activity of myeloperoxidase(MPO) in rats.RT-PCR real-time analysis,interleukin-1(IL-1),IL-6,tumor necrosis factor-α(TNF-α)mRNA expression.A constant phase model is used to assess lung mechanics assessment.Results At 6 h,12 h,and 24 h after FEDAC injection,the lung radioactivity absorption of the model group increased compared with the control group(P < 0.05).Compared with control rats,the model group stimulated alveolar congestion,and mainly increased the thickness of the alveolar wall and the infiltration or aggregation of inflammatory cells.Compared with the control group,the model group had a comprehensive score of inflammation [(6.84 ± 1.37) vs.(1.37 ± 0.26) points],reactive oxygen species(ROS) content [(4.19 ± 1.24) vs.(1.57 ± 0.23) nmol/L) ],and myeloperoxidase(MPO) activity [(6.31 ± 1.27)vs.(1.08 ± 0.14) nmol/L],the mRNA expression of inflammation indicators IL-1(1.86 ± 0.24 vs.0.94 ± 0.08),IL-6(1.95 ± 0.26 vs.1.05 ± 0.13),TNF-α(2.01 ± 0.35 vs.1.14 ± 0.14),and lung resistance(Rrs) [(2.15 ± 0.14) vs.(1.24 ± 0.05) cm H2O.s/mL],lung elasticity(Ers) [(41.67 ± 5.08) vs.(41.67 ± 5.08) cm H2O.s/mL],Newtonian resistance(Rn) [(1.74 ± 0.12) vs.(0.78 ± 0.08)cm H2O.s/mL] and tissue damping(Gtis) [(8.36 ± 1.29) vs.(4.29 ± 0.67) cm H2O.s/mL] were significantly increased(P < 0.05).Conclusion Changes in Golgi apparatus stress levels in bronchial pneumonia rats,increased expression of pro-inflammatory cytokines,increased alveolar wall thickness,and infiltration or aggregation of inflammatory cells,resulting in decreased lung function.PET is used to monitor the progress of lung inflammation Useful tools.
