锦带花乙酸乙酯提取物镇痛作用研究

【摘要】 为了研究锦带花(Weigela florida)乙酸乙酯提取物对小鼠的镇痛作用,将锦带花利用乙酸乙酯萃取制备锦带花乙酸乙酯提取物,采用热板法和扭体法疼痛动物模型,将雌雄兼用小鼠100只随机分为锦带花乙酸乙酯提取物2.00、1.00、0.50 g/kg剂量组,盐酸曲马多0.04 g/kg剂量组和生理盐水组,每组10只,灌胃给药。分别观察给药后不同时间点的小鼠痛阈值,或不同时间段的小鼠扭体反应数。根据痛阈值和扭体反应数的变化评判锦带花乙酸乙酯提取物的镇痛作用。结果显示,在热板法中,锦带花乙酸乙酯提取物1.00 g/kg剂量组给药后60 min和2.00 g/kg剂量组给药后30、45、60 min的痛阈值高于生理盐水组;1.00 g/kg剂量组给药后45、60 min和2.00 g/kg剂量组给药后15~60 min的痛阈值分别高于各自给药前痛阈值(P<0.05或P<0.01)。在扭体法中,锦带花乙酸乙酯提取物0.50 g/kg剂量组给药后21~30 min,>31~40 min和>41~50 min,1g/kg剂量组给药后31~40 min和41~50 min以及2.00 g/kg剂量组给药后11~50 min 4个时间段的扭体反应数分别比生理盐水组明显减少(P<0.05或P<0.01),扭体反应抑制率显著提高。由此得出结论:锦带花乙酸乙酯提取物对小鼠具有一定的镇痛作用,且以2.00 g/kg剂量的镇痛效果最佳。盐酸曲马多具有显著的镇痛作用。

【Abstract】 In order to study the analgesic effect of Weigela Florida ethyl acetate extract on mice,ethyl acetate extract of Weigela Florida was prepared by ethyl acetate extraction.The hot plate method and writhing method were used to establish the pain animal model.100 male and female mice were randomly divided into 2.00,1.00 and 0.50 g/kg group,tramadol hydrochloride with 0.04 g/kg group.10 rats in each group were given intragastric administration.The pain threshold of mice at different time points and the numbers of writhing reactions in different time periods were observed.The analgesic effect of ethyl acetate extract from broccoli was evaluated according to the changes of pain threshold and writhing reaction numbers.The results showed that in the hot plate method,the pain threshold of 1.00 g/kg ethyl acetate extract group at 60 min and 2.00 g/kg group at 30,45 and 60 min after administration was higher than that in normal saline group;the pain threshold of 1.00 g/kg ethyl acetate extract group at 45,60 min and 2.00 g/kg group at 15~60 min after administration was higher than that before administration respectively(P<0.05 or P<0.01).In the writhing method,the numbers of writhing reaction 0.50 g/kg of ethyl acetate extract group were significantly decreased in the four time periods that 21~30 min,31~40 min and 41~50 min,1.00 g/kg group in 31~40 min and 41~50 min,and 2.00 g/kg group in 10~50 min(P<0.05 or P<0.01),and the inhibition rates of writhing reaction were significantly increased.The conclusion is drawn that the ethyl acetate extract of broccoli has certain analgesic effect on mice,and the analgesic effect of 2.00 g/kg is the best.Tramadol hydrochloride has significant analgesic effect.
