聚酰胺纯化膜苞鸢尾中总黄酮的工艺研究

【摘要】 目的研究聚酰胺柱色谱纯化膜苞鸢尾中总黄酮的工艺条件。斱法以膜苞鸢尾中总黄酮的解析率与纯度为指标,通过考察静态、动态吸附试验,优化聚酰胺纯化膜苞鸢尾中总黄酮的工艺条件。结果聚酰胺对总黄酮有良好的纯化效果,其优化工艺为:最佳上柱样品溶液质量浓度2.0mg·mL-1,pH=3,吸附流速1 BV·h-1,吸附1.5 h后,70%乙醇洗脱液以1 BV·h-1速度洗脱。总黄酮动态解析率为92.34%,纯度45.59%。结论聚酰胺对膜苞鸢尾中总黄酮纯化性能较高,纯度提高了2.9倍。

【Abstract】 Objective: To optimize the purification method of total flavonoids from Iris scariosa by polyamide resin. Methods: Using the desorption ratio and purity of total flavonoids from Iris scariosa as the indices, the purification of total flavonoids by polyamide resin was optimized with static and dynamic adsorption tests. Result: The optimal adsorption conditions were as follow: the concentration of sample solution was 2.0 mg·mL-1, pH was 3 and the flow velocity of adsorption was 1 BV·h-1, the sample solution was rested for 1.5 h in the chromatographic column of polyamide resin. 70% ethanol was used to elute the total flavonoids at the flow rate of 1 BV·h-1. The dynamic desorption ratio was 92.34% and the purity of total flavonoids was 45.59%. Conclusion: The total flavonoids in Iris scariosa was effectively purified by polyamide resin under the optimal conditions with 2.9 times increase.
