137Cs-γ辐照对实验红鲫AFLP多态性的影响

【摘要】 目的观察137Cs-γ辐照对实验红鲫AFLP的变化,探讨不同剂量137Cs-γ辐照对实验红鲫DNA多态性的影响。方法根据前期实验结果,将实验红鲫C1HD系随机分为5组,10尾/组,即空白对照组、1/16 LD50、1/8 LD50、1/4LD50、1/2 LD504个辐照组,设置1.94 Gy、3.88 Gy、7.76 Gy、15.53 Gy4个辐照剂量组和一个空白对照组,用生物辐照仪对实验红鲫进行一次性辐照。提取辐照前和辐照后的实验红鲫血液DNA,经AFLP-PCR扩增,并进行多态性信息含量(PIC)、基因杂合度(H)、香农信息指数(I)分析。结果实验红鲫在137Cs-γ辐照处理前的PIC、H和I分别为45.20%、0.19和0.28,经137Cs-γ辐照处理后分别为72.27%、0.28和0.41,且遗传学指数升高。结论经137Cs-γ辐照处理后,实验红鲫的AFLP多态性信息含量增加,基因杂合度升高。AFLP标记实验红鲫C1HD系可用于监测核辐射污染。

【Abstract】 Objective To observe the AFLP changes of137 Cs-γ irradiation on the laboratory red crucian carp,and to explore the effect of different doses of γ-rays on the laboratory red crucian carp DNA polymorphism. Method According to the preliminary experimental results,laboratory red crucian carp C1 HD was randomly divided into 5 groups,10 tails/group,namely blank control group,1/16 LD50,1/8 LD50,1/4 LD50,1/2 LD50,4 irradiation groups of 1. 94 Gy,3. 88 Gy,7. 76 Gy,15. 53 Gy,and blank control group were set,and the laboratory red crucian carp was irradiated once and for all by using a biological irradiator. The blood DNA of the laboratory red crucian carp before and after irradiation was extracted and amplified by AFLP-PCR. The polymorphism information content( PIC),gene heterozygosity( H) and Shannon information index( I) was analyzed. Result Before the laboratory red crucian carp was treated with137 Cs-γ irradiation,the gene polymorphism information content( PIC),Nei’s gene diversity( H) and Shannon’s Information index( I) in normal laboratory red crucian carp is 45. 20%,0. 19 and 0. 28. After137 Cs-γ irradiation treatment,they are 72. 27%,0. 28 and 0. 41 respectively,which showed the genetics index rise. Conclusion After137 Cs-γ nuclear radiation treatment,PIC and H in laboratory red crucian carp are higher. AFLP marker in laboratory red crucian carp C1 HD can be used to monitor nuclear contamination.
