SHP-1在人类癌症中作用机制的研究
Research on the mechanism of SHP-1 in human cancer
【摘要】 蛋白质酪氨酸激酶(PTKs)和蛋白质酪氨酸磷酸酶(PTPs)的协调作用维持了细胞内酪氨酸磷酸化水平的平衡,对生长、分化、代谢、迁移和存活在内的多种细胞过程至关重要。酪氨酸磷酸化异常是PTKs和PTPs活性失衡的结果,与包括癌症在内的多种人类疾病的发病机制有关。SHP-1(Src homology region 2 domain-containing phosphatase 1)又称为PTPN6,是一种主要在造血细胞和上皮细胞中表达的非受体型蛋白质酪氨酸磷酸酶。SHP-1是一种调节炎症和免疫反应的重要蛋白,其通过对靶分子去磷酸化调控细胞反应。另外在很多癌症中也都发现了SHP-1蛋白表达的异常。本文全面的总结了SHP-1在癌症中的最新研究进展,主要从SHP-1的基本结构特征、在不同癌症中参与调控的信号通路与发挥的作用,以及现阶段针对于SHP-1的靶向药物进行阐述,旨在通过对现阶段SHP-1研究的总结,为未来SHP-1的研究提供新的思路。
【Abstract】 The coordination of protein tyrosine kinases(PTKs) and protein tyrosine phosphatases(PTPs) maintains the balance of intracellular tyrosine phosphorylation,which is essential for many cell processes including growth,differentiation,metabolism,migration and survival.The abnormal phosphorylation of tyrosine is the result of the imbalance of PTKs and PTPs activity,which is related to the pathogenesis of many human diseases,including cancer.SHP-1(Src homology region 2 domain-containing phosphatase 1),also known as PTPN6,is a non-receptor-like PTP mainly expressed in hematopoietic cells and epithelial cells.SHP-1 is an important protein regulating inflammation and immune response,which regulates signaling pathways by dephosphorylation of target molecules.In addition,abnormal expression of SHP-1 protein has been found in many cancers.This paper summarizes the latest research progress of SHP-1 in cancer comprehensively from the following aspects.The basic structural characteristics of SHP-1,the signaling pathway regulated by SHP-1,the role played in different cancers,as well as the new target drugs of SHP-1.We aim to provide new ideas for the future research of SHP-1 through the summary of the current stage of SHP-1 research.
