南方根结线虫侵染下罗汉果代谢组学研究
Metabonomics analysis of Siraitia grosvenorii under infection of Meloidogyne incognita
【摘要】 罗汉果为广西特有经济作物,但其主栽品种易受根结线虫的侵害,而对于罗汉果植株根系受根结线虫侵染早期其代谢变化情况知之甚少。基于LC-MS技术的代谢组学方法,对南方根结线虫侵染下的罗汉果根系进行了代谢轮廓变化非靶向分析。主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)结果表明,接种南方根结线虫7 d后,是否受到根结线虫侵染的罗汉果之间其根系代谢物呈现明显分离。单变量分析和t检验分析结果显示,有16种代谢物含量发生显著性差异变化,这些代谢物主要涉及糖代谢、脂类代谢和氨基酸代谢。结果可为进一步揭示南方根结线虫与罗汉果根系互作的分子机制提供参考和研究思路。
【Abstract】 Siraitia grosvenii is a special economic crop in Guangxi Autonomous Region and its main varieties are susceptible to rootknot nematodes,but the change of metabolites of Siraitia grosvenorii infected by M. incognita in the early stage was poorly understood.In this study,non-target analysis based on liquid chromatography-mass spectrometry was used to analyze the metabolic profile of the roots of Siraitia grosvenii seedlings infected by M. incognita. Principal component analysis and orthogonal partial least-squares discrimination analysis were also carried out for pattern recognition. The result showed that a clear separation was achieved between the control and treated with M. incognita at 7 days post-inoculate. Statistical significance was assessed with a t-test and a 2-fold change and 16 metabolites in root extracts of Siraitia grosvenii changed significantly in the level P < 0. 05. These metabolites were mainly related to glucose metabolism,lipid and amino acid metabolism. The result here can provide insight into interaction between Siraitia grosvenii and M. incognita in early phase.
