IL-37对氧化低密度脂蛋白诱导人冠状动脉内皮细胞分泌单核细胞趋化蛋白1的影响
Effect of interleukin-37 on the secretion of monocyte chemoattractant protein-1 in human coronary artery endothelial cells induced by oxidized-low density lipoprotein
【摘要】 目的:探讨白细胞介素37(IL-37)对氧化低密度脂蛋白(ox-LDL)诱导人冠状动脉内皮细胞(HCAECs)分泌单核细胞趋化蛋白1(MCP-1)的影响。方法:HCAECs经siIL-37 mRNA或IL-37b过表达质粒转染24 h后,用80μg/mL ox-LDL处理24h,同时设置空白对照组、未转染ox-LDL处理组、siRNA阴性对照组、空白质粒组。MTT法检测各组细胞增殖能力,RTPCR法检测各组细胞中MCP-1 mRNA的表达,ELISA法检测各组细胞培养液上清中MCP-1蛋白水平。结果:未转染oxLDL处理组细胞内MCP-1 mRNA的表达与培养液上清中的MCP-1蛋白的分泌量与对照组相比均有升高(P<0.05)。siIL-37mRNA组的MCP-1 m RNA和蛋白水平均较siRNA阴性对照组增加(P<0.01)。IL-37b过表达质粒组的MCP-1 mRNA和蛋白水平均较空白质粒组细胞减少(P<0.05)。结论:IL-37对ox-LDL诱导HCAECs分泌MCP-1有抑制作用。
【Abstract】 Objective: To investigate the effect of interleukin-37(IL-37) on the secretion of monocyte chemoattractant protein-1(MCP-1) in human coronary artery endothelial cells(HCAECs) induced by oxidizedlow density lipoprotein(ox-LDL). Methods: HCAECs were transfected with siIL-37 mRNA or IL-37 b overexpression plasmid for 24 h, and then treated with 80 μg/mL ox-LDL for 24 h. At the same time, blank control group, non transfected ox-LDL treatment group, siRNA negative control group and blank plasmid group were set up. MTT assay was used to detect cell proliferation, RT-PCR was used to detect MCP-1 mRNA expression, and ELISA method was used to detect MCP-1 protein level in cell culture supernatant. Results: Compared with the control group, the expression of MCP-1 mRNA and the secretion of MCP-1 protein in the supernatant of culture medium in the non transfected ox-LDL group were significantly higher than those in the control group(P<0.05).The mRNA and protein levels of MCP-1 in the siRNA-37 mRNA group were higher than those in the siRNA lnoewgearti vthe acno ntthroosl eg rionu bpl(aPnk< 0 p.l0 a1 s).m iMd CgPr-ou1 p m(RPN<0 A. 0 a5 n).d Cproontecilnu slieovne:l sI iLn-I3 L7-3 ca7 nb ionvheirbeixtp trheses isoenc rpeltiaosnm iodf gMroCuPp-w1 e irne HCAECs induced by ox-LDL.
