快速眼动睡眠剥夺对抑郁模型大鼠海马区胶质细胞源性神经营养因子的影响

【摘要】 目的:探讨快速眼动睡眠剥夺(REMSD)对抑郁模型大鼠海马区胶质细胞源性神经营养因子(GDNF)的影响和REMSD快速抗抑郁的可能机制。方法:18只雄性SD大鼠随机分为正常对照组、应激造模组和REMSD组,每组6只。使用慢性不可预知温和应激(CUMS)法建立抑郁大鼠模型,通过小平台水环境法连续剥夺72 h大鼠的快速眼动睡眠。采用开场实验检测大鼠的自发活动,蛋白质印迹法检测大鼠海马区GDNF的表达水平。结果:应激造模组、REMSD组大鼠在CUMS 28 d后,自发活动开场实验中总路程和中央路程较正常对照组均减少(P<0.01)。REMSD组大鼠在睡眠剥夺72 h后,自发活动开场实验中总路程和中央路程较睡眠剥夺前增加(P<0.01)。应激造模组及REMSD组大鼠海马区GDNF的表达较正常对照组均降低(P<0.05);但REMSD组与应激造模组GDNF的表达差异无统计学意义(P>0.05)。结论:REMSD可以快速改善大鼠的抑郁样行为,但GDNF本身可能并未参与快速抗抑郁的机制。

【Abstract】 Objective:To investigate the effect of rapid eye movement sleep deprivation(REMSD) on glial cell line-derived neurotrophic factor(GDNF) in hippocampus of depression model rats and the possible mechanism of rapid antidepressant of REMSD. Methods:18 male Sprague Dawley rats were randomly divided into normal control group,stress model group and REMSD group. The depression rat model was established by chronic unpredictable mild stress(CUMS). The rapid eye movement sleep was deprived by small platform water environment method for 72 hours. The spontaneous activity of rats was detected by open field test,and the expression of GDNF in hippocampus was detected by Western blotting. Results:After 28 days of CUMS,the total distance and the central distance in the open field test of spontaneous activity of rats in the stress model group and the REMSD group decreased compared with the normal control group(P<0.01). After 72 hours of sleep deprivation,the total distance and central distance in the open field test of spontaneous activity in rats in the REMSD group increased compared with those before sleep deprivation(P<0.01). The expression of GDNF in hippocampus of rats in stress group and REMSD group was lower than that in normal control group(P<0.05),but there was no significant difference between the two groups(P>0.05). Conclusion:REMSD can improve the depressive behavior of rats rapidly,but GDNF itself may not be involved in the mechanism of rapid antidepressant.
