脑梗死患者骨保护素基因多态性与血浆中骨保护素水平的关系
ASSOCIATION OF OSTEOPROTEGERIN GENE POLYMORPHISM WITH PLASMA OSTEOPROTEGERIN LEVEL IN PATIENTS WITH CEREBRAL INFARCTION
【摘要】 目的探讨大动脉粥样硬化(LAA)型脑梗死患者骨保护素(OPG)基因多态性与血浆中OPG水平的关系。方法收集LAA型脑梗死患者(LAA组)221例及健康对照者(对照组)115例,采用酶联免疫吸附测定法(ELISA)检测受检者血浆中OPG水平;应用聚合酶链反应限制性片段长度多态性(PCR-RFLP)技术检测受检者OPG基因rs3102735位点多态性。结果 LAA组血浆中OPG水平高于对照组(t=4.826,P<0.05);LAA组OPG基因rs3102735位点CC/CT基因型及等位基因C频率显著高于对照组(χ~2=6.408、8.903,P<0.05);两者与缺血性卒中的发生显著相关(OR=1.574,95%CI=1.063~3.154,P<0.05;OR=1.633,95%CI=1.182~2.665,P<0.05)。携带CC/CT基因型及等位基因C患者血浆中OPG水平显著高于对照组(χ~2=5.377、6.872,P<0.05);CC/CT基因型及C等位基因与OPG高水平有关(OR=1.002,95%CI=1.001~1.0003,P<0.05,OR=1.572,95%CI=1.019~2.425,P<0.05)。结论 LAA脑梗死患者血浆中OPG水平增高,OPG基因rs3102735位点C等位基因及CC/CT基因型可能与血浆高OPG水平有关。
【Abstract】 Objective To investigate the association of osteoprotegerin(OPG) gene polymorphism with plasma OPG level in patients with large-artery atherosclerosis(LAA) cerebral infarction. Methods A total of 221 patients with LAA cerebral infarction were enrolled as LAA group, and 115 healthy controls were enrolled as control group. ELISA was used to measure the plasma level of OPG, and polymerase chain reaction-restriction fragment length polymorphism was used to detect the polymorphism of OPG rs3102735. Results The LAA group had a significantly higher plasma OPG level than the control group(t=4.826,P<0.05). Compared with the control group, the LAA group had significantly higher frequencies of CC/CT genotype and C allele of OPG rs3102735(χ~2=6.408,8.903,P<0.05), which were significantly correlated with the development of ischemic stroke(OR=1.574,95%CI=1.063-3.154,P<0.05;OR=1.633,95%CI=1.182-2.665,P<0.05). The patients carrying CC/CT genotype and C allele had a significantly higher plasma OPG level than the control group(χ~2=5.377,6.872,P<0.05), and CC/CT ge-notype(OR=1.002,95%CI=1.001-1.0003,P<0.05) and C allele(OR=1.572,95%CI=1.019-2.425,P<0.05) were associa-ted with high OPG level. Conclusion Patients with LAA cerebral infarction have an increased plasma OPG level, and C allele and CC/CT genotype at OPG rs3102735 may be associated with the high plasma level of OPG.
