高压氧对2型糖尿病小鼠摄食及胰岛素敏感性的影响

【摘要】 目的探讨高压氧(HBO)对2型糖尿病(2DM)小鼠摄食水平、下丘脑弓状核神经肽Y的表达和胰岛素抵抗的影响。方法雄性C57BL/6J小鼠腹腔注射链脲佐菌素,并给予高脂饮食(HFD)喂养诱导2DM。年龄相同的小鼠单纯喂食HFD作为对照组。造模成功的小鼠随机分为2DM组、2DM+HBO组,HFD的小鼠随机分为HFD组、HFD+HBO组。HFD+HBO组和2DM+HBO组小鼠给予2个绝对大气压、体积分数1.00的氧气持续1周的治疗,HFD组和2DM组小鼠置于相同环境不做处理,治疗结束后观察各组小鼠空腹血糖、胰岛素抵抗指数和摄食量的变化,采用免疫荧光法检测各组小鼠下丘脑弓状核中神经肽Y的表达量。结果与2DM组相比较,2DM+HBO组小鼠空腹血糖水平以及胰岛素抵抗指数显著降低(F=11.206、36.567,P<0.05),摄食量显著增加(F=7.598,P<0.05)。与HFD组相比,HFD+HBO组小鼠空腹血糖、胰岛素抵抗指数和摄食量并未明显改变。下丘脑弓状核中神经肽Y的表达呈现出与摄食水平相一致的结果。结论高压氧可增加2DM小鼠摄食量及下丘脑弓状核中神经肽Y的阳性神经元数量,同时还可改善胰岛素敏感性。

【Abstract】 Objective To investigate effects of hyperbaric oxygen(HBO) on food intake, the expression of neuropeptide Y in the hypothalamic arcuate nucleus, and insulin resistance in type 2 diabetic mice. Methods Male C57BL/6J mice were injected intraperitoneally with streptozotocin and fed with a high-fat diet(HFD) to induce type 2 diabetes mellitus(2 DM). The age-matched mice fed with HFD only were selected as control group. The successfully induced diabetic mice were randomly divided into 2 DM group and 2 DM+HBO group, while the mice in the control group were randomly divided into HFD group and HFD+HBO group. The mice in the HFD+HBO group and 2 DM+HBO group were treated with hyperbaric oxygen(2 absolute atmospheres, 100% vol. oxygen) for one week. The mice in the HFD group and 2 DM group were placed in the same environment wi-thout treatment. At the end of the experiment, the fasting blood glucose, insulin resistance index, and food intake of the mice in each group were observed. The expression of neuropeptide Y in the hypothalamic arcuate nucleus of the mice in each group was measured by the immunofluorescence staining technique. Results Compared with the 2 DM group, the 2 DM+HBO group had significantly reduced fasting blood glucose and insulin resistance index(F=11.206,36.567,P<0.05) and significantly increased food intake(F=7.598,P<0.05). Compared with the HFD group, the HFD+HBO group showed no significant changes in fasting blood glucose, insulin resistance index, and food intake. The expression of neuropeptide Y in the hypothalamic arcuate nucleus was consis-tent with the level of food intake. Conclusion HBO increases food intake and the number of neuropeptide Y-po-sitive neurons in the hypothalamic arcuate nucleus and improves insulin sensitivity in type 2 diabetic mice.
