食管癌组织中miR-1269a、SOX6的表达变化及意义
Expression changes of miR-1269a and SOX6 in esophageal carcinoma tissues and their clinical significance
【摘要】 目的观察食管癌组织中微小RNA(miRNA)-1269a及转录因子SOX6的表达变化,并探讨其意义。方法采用实时荧光定量PCR(qRT-PCR)法检测120例份食管癌组织和癌旁正常组织中miR-1269a及SOX6的表达,分析miR-1269a、SOX6的表达与患者临床病理特征的关系。结果食管癌组织中SOX6相对表达量低于癌旁正常组织,而食管癌组织中miR-1269a相对表达量高于癌旁正常组织(P均<0. 05)。癌组织中miR-1269a与SOX6的表达呈负相关(r=-0. 716,P=0. 000)。食管癌组织中miR-1269a、SOX6表达与肿瘤TNM分期、组织分化程度及肿瘤浸润深度有关(P均<0. 05),与患者年龄、性别及淋巴结转移无关(P均> 0. 05)。结论食管癌组织中miR-1269a表达上调,SOX6表达下调;二者联合检测有助于食管癌病情判断。
【Abstract】 Objective To observe the expression levels of miR-1269 a and transcription factor SOX6 in the esophageal cancer tissues,and to analyze its significance. Methods The expression levels of miR-1269 a and SOX6 in 120 cases of esophageal cancer tissues and adjacent normal tissues were detected by real-time quantitative PCR( qRT-PCR),the relationships between miR-1269 a and SOX6 expression differences and clinicopathological characteristics were analyzed. Results The expression of SOX6 in the esophageal cancer tissues was lower than that in the adjacent normal tissues,while the expression of miR-1269 a in the esophageal cancer was higher than that in the adjacent normal tissues,with statistically significant differences( both P < 0. 05). The expression of miR-1269 a and SOX6 in the cancer tissues was negatively correlated( r =-0. 716,P = 0. 000). The expression levels of miR-1269 a and SOX6 in the esophageal cancer were related to tumor TNM stage,differentiation degree,and tumor invasion depth( all P < 0. 05),but not to age,gender or whether there was lymph node metastasis( all P > 0. 05). Conclusion The expression of miR-1269 a is up-regulated and the expression of SOX6 is down-regulated in esophageal cancer,which is related to the TNM stage,differentiation degree and tumor invasion depth of the tumor; the combined detection of the two is helpful for the judgment of esophageal cancer.
