胃癌组织中miR-200a-3p、G3BP2的表达及意义
Expression and significance of miR-200a-3p and G3BP2 in gastric cancer tissues
【摘要】 目的研究胃癌组织中微小RNA(miRNA)-200a-3p及GTP酶激活蛋白SH3功能区结合蛋白2(G3BP2)表达及意义。方法应用qRT-PCR法检测97例份胃癌组织及其配对的癌旁正常组织(距离癌组织5cm)中miR-200a-3p及G3BP2表达,分析miR-200a-3p、G3BP2表达差异及其与临床病理特征和预后的关系。结果与癌旁正常组织相比,胃癌组织中miR-200a-3p表达降低,而G3BP2表达升高(P均<0. 05)。胃癌组织中miR-200a-3p表达与G3BP2呈显著负相关(r=-0. 610,P=0. 008)。胃癌组织中miR-200a-3p、G3BP2表达与肿瘤TNM分期、肿瘤分化有关(P均<0. 05),而与患者性别、年龄、肿瘤大小、位置、浸润深度及淋巴结转移无关(P均>0. 05)。miR-200a-3p低表达者3年总体生存率(OS)低于miR-200a-3p高表达者(P <0. 05); G3BP2高表达者3年OS低于G3BP2低表达者(P <0. 05)。结论胃癌组织中miR-200a-3p表达降低,而G3BP2表达升高;二者联合检测有助于患者病情判断及预后评估。
【Abstract】 Objective To study the expression and clinical significance of microRNA(miR)-200 a-3p and GTPase activating protein SH3 domain binding protein 2(G3BP2) in the gastric cancer tissues. Methods The expression of miR-200a-3p and G3BP2 in 97 cases of gastric cancer tissues and its paracancer tissues was detected by real-time quantitative PCR(qRT-PCR). The differencial expression of G3BP2 and miR-200a-3p and their relationships with clinicopathological features and prognosis were analyzed statistically. Results Compared with the paracancer tissues,the expression of miR-200a-3p was significantly lower in the gastric cancer tissues,while G3BP2 was significantly higher(both P < 0. 05).There was a significant negative correlation between miR-200a-3p and G3BP2 in the cancer tissues(r =-0. 634,P =0. 000). The expression of miR-200a-3p and G3BP2 in the cancer tissues was related to tumor TNM stage and tumor differentiation(all P < 0. 05),but not related to gender,age,tumor size,location,infiltration depth or lymph node metastasis(all P >0. 05). The 3-year overall survival rate(OS) of patients with low miR-200a-3p expression was significantly lower than that of patients with high miR-200a-3p expression(P < 0. 05). The 3-year OS of patients with high G3BP2 expression was significantly lower than that of patients with low G3BP2 expression(P < 0. 05). Conclusion The expression of miR-200a-3p decreases in the gastric cancer tissues,while the expression of G3BP2 is elevated; the combined detection of them is helpful for the disease judgment and prognosis evaluation.
