三氯生对雄斑马鱼鳃抗氧化相关基因表达的影响
Effect of Triclosan on Expression of Genes Related to Antioxidant in Gill of Male Zebrafish Danio rerio
【摘要】 为了探究三氯生(TCS)对硬骨鱼类鳃组织抗氧化能力影响的分子机制,采用半静态水体接触染毒法,将体长为1.5~2.1 cm的斑马鱼Danio rerio幼鱼分别暴露在0μg·L-1(对照组)、17.0μg·L-1、34.0μg·L-1和68.0μg·L-1TCS溶液中42 d后,利用普通PCR和RT-qPCR技术检测雄斑马鱼鳃组织抗氧化相关基因的表达水平。结果表明:在三种浓度TCS处理中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GPx)及金属硫蛋白-2(MT-2)基因表达量均下调,GPx1a基因的表达水平在68.0μg·L-1TCS浓度组显著下调(P<0.05),SOD、CAT和sMT-B基因的表达水平在34.0μg·L-1和68.0μg·L-1TCS组极显著下调(P<0.01),MT-2基因在各TCS浓度组中均表现极显著下调(P<0.01)。本研究结果表明,TCS抑制了雄斑马鱼鳃组织中抗氧化相关基因的表达,对雄斑马鱼鳃组织抗氧化防御系统造成了一定程度的损伤。
【Abstract】 Juvenile zebrafish(Danio rerio)with body length of 1.5 ~ 2.1 cm was exposed to various concentrations of triclosan(TCS)(0 μg·L-1, 17.0μg·L-1, 34.0 μg·L-1 and 68.0 μg·L-1)for 42 d and the expression of genes related to antioxidant in gill of the male zebrafish were investigated by semi-static water exposure method and by normal PCR and RT-qPCR techniques in order to explore antioxidant defense molecular mechanism of triclosan on bony fish gill tissue. It is showed that the expression levels of genes of superoxide dismutase(SOD), catalase(CAT), glutathione peroxidase(GPX)and metallothionein-2(MT-2)were down-regulated in TCS treated groups compared with those in the control group, with significantly down-regulated GPx1 a gene expression in the 68.0 μg·L-1 group(P < 0.05), very significantly down-regulated expression levels of SOD, CAT and sMT-B genes in the 34.0 μg·L-1 and 68.0 μg·L-1 groups(P < 0.01), and significantly down-regulated MT-2 gene in three different TCS concentrations groups(P < 0.01). The findings indicate that TCS inhibits m RNAs expression of antioxidant related genes, and can cause some degree of damage to the antioxidant defense system in gill tissues of male zebrafish.
