Zn-MT对运动大鼠心肌抗氧化酶及细胞凋亡蛋白表达量的影响
Effects of Zn-MT on the Expression of Antioxidant Enzymes and Apoptotic Proteins in Myocardium of Exercise Rats
【摘要】 目的:探讨锌-金属硫蛋白(Zn-MT)对力竭运动大鼠(♂)心肌细胞抗氧化酶、细胞凋亡率及Bax和Bcl-2蛋白表达量的影响。方法:选取雄性SD大鼠24只,随机分为运动对照组(A)、补充Zn-MT+运动组(B),两组大鼠均进行跑台运动。于力竭运动后即刻取材,抗氧化酶采用比色法,心肌凋亡细胞检测采用TUNEL法,Bcl-2、Bax蛋白表达检测采用免疫组化法。结果:C组和B组相比,C组GSH-Px、CAT、GSH、SOD活性有不同程度的升高;可以明显降低心肌细胞凋亡率和Bax蛋白表达率,Bcl-2表达明显增高。结论:补充Zn-MT可以MT可以提高心肌抗氧化酶活性,通过影响心肌细胞中Bcl-2和Bax蛋白表达,减少力竭运动后的心肌细胞凋亡。
【Abstract】 Purpose: Probe into the influence of myocardium cell anti-oxidase, apoptosis、Bax、Bcl-2 about Zn-MT to exhaust mouse. Method:Choose 24 male SD big mice, assign to the sport control group(A), supplementing Zn-MT+sport group(B)at random. B,C groups draw materials after strength exhaust sports,adopt the colorimetry to detect anti-oxidase,adopt TUNEL law to detect myocardium apoptosis、Bcl-2, adopt immune group law detect Bax albumen express. Result:In B Group, GSH-Px, CAT, GSH, SOD active decline, there is rising after supplementing Zn-MT,compared with group B, C group myocardium apoptosis and Bax protein express decrease obviously,Bcl-2 expresses increases. Conclusion: supplementing Zn-MT Can improve the myocardium anti-oxidase activity, reduce the myocardium cells after strength exhausts sports and apoptosis through influencing the myocardium.
