大肠杆菌K88ac-ST1-LTB融合蛋白多价卵黄抗体的制备

【摘要】 对已构建的重组菌株BL21(DE3)(pETXKSL3)进行鉴定,酶切鉴定证实重组质粒pETXKSL3含有K88ac-ST1-LTB融合基因且阅读框架正确.Western blot分析表明K88ac-ST1-LTB融合蛋白能够被ST1单抗、K88ac和LTB抗体识别.以K88ac-ST1-LTB融合蛋白为抗原免疫蛋鸡制备卵黄抗体,ELISA法测定卵黄抗体效价可达1∶13 200.仔猪攻毒实验表明制备的卵黄抗体对仔猪黄痢和仔猪白痢的预防有效率为96%,上述研究结果说明K88ac-ST1-LTB多价卵黄抗体对由ETEC引起的新生仔猪腹泻具有很好的预防效果,从而为最终开发出一种新型高效、可替代抗生素的防治新生仔猪大肠杆菌性腹泻的特异性卵黄抗体生物防治制剂打下坚实基础.

【Abstract】 By identifying the BL21(DE3)(pETXKSL3)recombinant strain was identified and restriction endonuclease digestion confirmed that the pETXKSL3 recombinant plasmid contained the K88ac-ST1-LTB fusion gene and the reading frame was correct.Western blot analysis showed that the K88ac-ST1-LTB fusion protein was recognized by ST1mAb,K88ac and LTB antibodies.Egg yolk antibody was prepared by immunizing hens with K88ac-ST1-LTB fusion protein as antigen.The titer of yolk antibody can reach 1∶13 200 by ELISA.Piglet challenge experiments showed that the effective rate of the prepared egg yolk antibody was 96% against yellow scours of newborn piglets and white scours of piglets.The above results indicate that K88ac-ST1-LTB multivalent yolk antibody has a good preventive effect on newborn piglet diarrhea caused by ETEC.Therefore,it has laid a solid foundation for the development of a high-efficiency alternative antibiotic yolk antibody which can prevent the newborn piglet diarrhea caused by ETEC.
