隐丹参酮联合顺铂对非小细胞肺癌PC9细胞MMP-9、MMP-2、p-ERK1/2表达和迁移侵袭的影响
Effect of cryptotanshinone combined with cisplatin on MMP-9,MMP-2,p-ERK1/2 expressions and migration and invasion in non-small cell lung cancer PC9 cell
【摘要】 目的研究隐丹参酮(CPT)联合顺铂(DDP)作用于肺癌细胞PC9细胞后,基质金属蛋白酶2(MMP-2)、基质金属蛋白酶9(MMP-9)和磷酸化细胞外信号调节激酶1/2(p-ERK1/2)的表达及其对细胞生长迁移能力的影响。方法体外培养PC9细胞细胞株,分为空白对照组、CPT组、DDP组及CPT联合DDP组,CCK-8法检测各组PC9细胞的增殖情况,免疫印迹技术检测MMP-2、MMP-9、p-ERK1/2蛋白的表达水平,RT-PCR检测MMP-9及MMP-2的mRNA转录水平,Trasswell技术检测用药后PC9细胞的穿透滤膜数。结果与空白对照组、5μg/ml DDP、10μmol/L CPT相比,5μg/ml DDP+10μmol/L CPT组细胞存活率明显降低(t分别=11.82、9.00、7.88,P均<0.05),MMP-9、MMP-2蛋白表达水平明显下调(t分别=9.82、8.02、7.83;13.66、1.76、7.40,P均<0.05),p-ERK1/2蛋白表达水平明显下降(t分别=11.50、17.75、10.72,P均<0.05),PC9细胞穿透滤膜数目明显减少,迁移能力降低。结论CPT联合DDP可通过下调MMP-2、MMP-9、p-ERK1/2的表达,抑制非小细胞肺癌细胞的迁移和侵袭。
【Abstract】 Objective To investigate the expressions of MMP-2,MMP-9 and p-ERK1/2,the effects on cell growth,migration and invasion after NSCLC PC9 cell stimulated by cryptotanshinone combined with cisplatin. Methods PC9 cells were divided into control group,cryptotanshinone group,cisplatin group and cryptotanshinone combined cisplatin group.The proliferation of PC9 cells was determined by CCK-8 assay,the levels of MMP-2,MMP-9,p-ERK1/2 were detected by Western blot,the mRNA level MMP-2 and MMP-9 were detected by RT-PCR,the number of penetrating filter membranes of PC9 was detected by Trasswell technology. Results Compared to the control group,5 μg/ml cisplatin group,and 10 μmol/L cryptotanshinone group,the survival rate of cell proliferation of cryptotanshinone combined cisplatin group was significantly lower(t=11.82,9.00,7.88,P<0.05),the expressions of MMP-2,MMP-9,p-ERK1/2 were significantly lower(t=9.82,8.02,7.83,13.66,1.76,7.40,11.50,17.75,10.72,P<0.05),and the number of cells penetrating the membrane was significantly decreasing. Conclusion Cryptotanshinone combined with cisplatin can inhibit the migration and invasion of NSCLC cells by down-regulating the expressions of MMP-2,MMP-9,and p-ERK1/2.
