正交设计优化富硒甘薯中硒蛋白提取方法
Orthogonal Design Optimization of Selenium Protein Extraction Method from Selenium-Enriched Sweet Potato
【摘要】 将浸提剂种类、浸提剂浓度、料液比、浸提时间、浸提温度单因素实验结合四因素三水平正交试验优化富硒甘薯中可溶性蛋白提取工艺。结果发现富硒甘薯中可溶性蛋白最佳浸提条件为:浓度为0.15 mol/L的NaOH溶液作为浸提剂,料液比1∶30,40℃恒温水浴浸提90 min。此条件下0.5 g富硒甘薯冻干粉可溶性蛋白得率为4.6%;蛋白中硒含量为1.27μg/L,硒得率为34.4%。优化后的实验工艺操作简单、蛋白得率高,可用于推广富硒薯类农作物功能性产业开发。
【Abstract】 The type of extractant, extractant concentration, material-to-liquid ratio, extraction time, and extraction temperature were combined with a four-factor three-level orthogonal test to optimize the extraction process of soluble protein in selenium-enriched sweet potato. The results showed that the optimal extraction conditions for soluble protein in selenium-enriched sweet potato were as follows: Na OH solution with a concentration of 0.15 mol/L was used as the extractant, the ratio of material to liquid was 1:30, and the extraction was performed in a constant temperature water bath at 40 ℃ for 90 min. Under these conditions, the soluble protein yield of 0.5 g selenium-enriched sweet potato freeze-dried powder was 4.6%. The selenium content in the protein was 1.27 μg/L, and the selenium yield was 34.4%. The optimized experimental technology has simple operation and high protein yield, and can be used to promote the development of functional industries of selenium-rich potato crops.
