河南奶山羊线粒体D-loop区遗传多态性和遗传分化研究

Genetic diversity and genetic differentiation analysis of mitochondrion D-loop region in Henan dairy goats

【摘要】 为了解河南奶山羊遗传资源现状,对河南奶山羊、萨能奶山羊、关中奶山羊和伏牛白山羊线粒体DNA D-loop区全序列遗传多态性和遗传关系进行分析。结果表明,共存在16种单倍型,各品种单倍型多样度范围为0.391~0.814,核苷酸多样度的范围为0.000 35~0.004 64,核苷酸平均差异数范围为9.333 33~13.666 67,核苷酸岐异度变异范围为0.008 33~0.012 19,遗传分化系数范围为0.135 65~0.360 10,固定系数范围为0.583 23~0.935 50。山羊品种间系统发育树表明,河南奶山羊首先与伏牛白山羊聚为一支,其次河南奶山羊与萨能奶山羊聚类,最后与关中奶山羊聚类。系统发育树聚类结果与遗传距离分析、遗传分化分析结果相符,从遗传角度证实了河南奶山羊是以萨能奶山羊为父本,以河南地方品种为母本经杂交选育而成。

【Abstract】 To understand the current situation of the genetic resources of Henan dairy goats the genetic diversity of mitochondrion(mt DNA) D-loop and genetic relationship of Henan dairy goats, Saanen dairy goats, Guanzhong dairy goats, and Funiu white dairy goats were analyzed in this study. The results showed 16 haplotypes were identified based on D-loop variations. The haplotype diversity ranged from 0.391 to 0.814, the nucleotide diversity ranged from 0.000 35 to 0.004 64, the mean nucleotide difference ranged from 9.333 33 to 13.666 67, the nucleotide divergence variation ranged from 0.008 33 to 0.012 19, the genetic differentiation coefficient ranged from 0.135 65 to 0.360 10, and the fixed coefficient ranged from 0.583 23 to 0.935 50. The phylogenetic tree showed that Henan dairy goats clustered with Funiu white goats firstly, then they got together with Sannen dairy goats, and finally, they clustered with Guanzhong dairy goats. The results of phylogenetic tree corresponded with those of genetic distance and genetic differentiation analysis, verifying that Henan dairy goats were hybridized from Saanen as paternal line and domestic goats of Henan Province as maternal line.
