橙色红曲菌Rab蛋白家族的生物信息学分析
Bioinformatics analysis of the Rab proteins in Monascus aurantiacus
【摘要】 Rab蛋白调控蛋白在细胞中的运输,发现其与丝状真菌的生长发育、液泡融合、自噬及次级代谢产物的合成有关。为鉴定及分析橙色红曲菌(Monascus aurantiacus)AS3.4384中的Rab蛋白家族种类及功能,本论文以酵母菌Rab家族的蛋白序列为同源序列,利用Blast程序在橙色红曲菌蛋白序列中进行同源搜索,并利用生物学软件对所预测到的Rab蛋白进行性质预测、结构域分析、基因结构分析和系统进化分析。结果发现,AS3.4384中共有9个Rab蛋白,均为小分子量的亲水蛋白且均能定位于细胞器膜上。通过系统发育树及序列比对分析,发现此9个Rab蛋白在红曲菌中是相对保守的。本研究为具体深入研究Rab蛋白的功能提供理论依据,为减少红曲菌产生桔霉素提供新思路。
【Abstract】 The Rab family regulates the transport of proteins in cells.It is found that Rab is involved in the growth and development of filamentous fungi,vacuole fusion,autophagy and the synthesis of secondary metabolites.Using Blast program and the protein sequence of the yeast Rab family as homologous sequence,Rab proteins were searched in the protein sequence of Monascus Aurantiacus and the diversification and function of the Rab proteins family were identified and analyzed from M aurantiacus AS3.4384.Then their property prediction,structural domains,gene structure,and phylogeny were analyzed.The results showed that there were 9 Rab proteins in AS3.4384,all of which were small molecular hydrophilic proteins and localized on the orgnnelle membrane.Phylogenetic tree and sequence alignment of these proteins indicated the Rab proteins are relatively conserved in M.aurantiacus.This study provides a theoretical basis for studying the function of the Rab proteins in Monascus and may develop a new idea for reducing the production of citrinin in Monascus.
