淫羊藿苷对糖尿病肾病大鼠肾功能的保护作用及相关机制
Protective effects of icariin on renal function in rats with diabetic nephropathy and the related mechanisms
【摘要】 目的研究淫羊藿苷对糖尿病肾病大鼠肾功能的保护,并初步分析其作用机制。方法随机将40只SD大鼠分为正常组(Normal)、模型组(DN)、阳性对照组(厄贝沙坦IRB:25 mg/kg)、ICA(淫羊藿苷:100 mg/kg)组,每组10只,采用高糖高脂饮食加腹腔注射链脲霉素构建糖尿病肾病(DN)模型,检测肾功能指标:尿白蛋白(UAER)、血肌酐(Scr)、血尿素氮(BUN)。通过HE染色观察大鼠肾组织病理学形态变化;ELISA检测大鼠血清中IL-1β、IL-6、MCP-1和肾组织中SOD、GSH-px和MDA含量;免疫组化检测大鼠肾组织中Caspase-3的表达;Western blot检测大鼠肾组织TGF-β、VEGF、p-p65的表达。结果与Normal组相比,DN组大鼠UAER、Scr、BUN、MDA、IL-1β、IL-6、MCP-1、Caspase-3、TGF-β、VEGF、p-p65表达显著上升,SOD、GSH-px表达显著下降(P<0.05);给予ICA处理后,大鼠UAER、Scr、BUN、MDA、IL-1β、IL-6、MCP-1、Caspase-3、TGF-β、VEGF、p-p65表达显著下降,SOD及GSH-px表达显著上升(P<0.05)。结论淫羊藿苷对糖尿病肾病大鼠肾功能有一定的保护作用,可能减轻大鼠炎症反应、氧化应激水平,减少细胞外基质的增生并抑制肾组织细胞的凋亡,从而减少肾组织的损伤。
【Abstract】 This study was designed to evaluate the protective effects of icariin on renal function in rats with diabetic nephropathy and to explore the mechanism preliminarily. Total of 40 SD rats were randomly divided into normal group(normal), model group(DN), positive control group(irbesartan IRB, 25 mg/kg) and ICA(icianin, 100 mg/kg) goup, with 10 rats in each group. High-glucose and high-fat diet + intraperitoneal injection of streptozotocin were used to construct diabetic nephropathy(DN) model, and the renal function indicators(urinary albumin excretion rate(UAER), serum creatinine(Scr), blood urea nitrogen(BUN)) were detected. HE staining was used to observe the pathological morphological changes of renal tissues; ELISA was applied to detect the levels of IL-1β,IL-6 and MCP-1 in serum and the levels of SOD, GSH-px and MDA in renal tissues. The expression level of Caspase-3 in renal tissues was detected by immunohistochemistry, and the expression levels of TGF-β, VEGF and p-p65 in renal tissues were detected by Western blotting. Compared with normal group, the expression levels of UAER, Scr, BUN, MDA, IL-1β, IL-6, MCP-1, Caspase-3, TGF-β, VEGF and p-p65 in DN group were increased significantly, while the expression levels of SOD and GSH-px were decreased significantly. After ICA treatment, the expression levels of UAER, Scr, BUN, MDA, IL-1β, IL-6, MCP-1, Caspase-3, TGF-β, VEGF and p-p65 were significantly decreased, while the expression levels of SOD and GSH-px were significantly increased. In conclusion,icariin has a certain protective effect on renal function in rats with diabetic nephropathy. And it may reduce the renal tissue damage by relieving inflammatory response and oxidative stress, reducing the proliferation of extracellular matrix and inhibiting the apoptosis of renal tissue cells.
