微滴式数字PCR技术在新型冠状病毒检测中的潜在价值
Potential value of droplet digital PCR in detection of SARS-CoV-2
【摘要】 采用荧光定量PCR (RT-qPCR)检测样本中的新型冠状病毒核酸阳性率低,导致得多次反复检测。假阴性的存在使得部分携带者漏诊,成为传染疾病的巨大隐患,尤其是当样本病毒核酸含量低、PCR抑制剂干扰、病毒发生变异与引物、探针结合能力下降等因素存在时。因此,亟待研究更高效的检测手段,防止超级传播者出现。本研究通过对应用微滴式数字PCR技术检测新型冠状病毒方法的探讨,为临床快速、准确诊断新型冠状病毒提供方案。
【Abstract】 Fluorescence quantitative PCR(RT-qPCR) is currently used to test viral nucleic acids in samples.However, in some cases, such as the low content of viral nucleic acids in samples, interference of PCR inhibitors and variation of viruses, there will still be false negative results leading to missed diagnosis. Therefore, it is urgent to develop more effective methods to detect new coronaviruses. We explored the application prospects of droplet digital PCR(ddPCR) in such detection, in order to seek a more efficient and sensitive detecting method to provide a rapid and accurate clinical diagnosis of SARS-CoV-2.
