酸性稻田土n-damo细菌的多样性和丰度

【摘要】 亚硝酸盐依赖型甲烷厌氧氧化(n-damo)细菌是新近发现的一类具有特殊功能的微生物,在废水深度脱氮领域具有潜在应用前景。研究利用n-damo菌16S rRNA和pmo A基因的专一性引物,通过系统发育分析和定量PCR技术,检测酸性稻田土中该类群的多样性和丰度。结果显示,在弱酸性中等养分水平的稻田土中,n-damo菌聚为3~4个OTUs类群;系统发育分析表明,酸性稻田土中存在新型未知n-damo类群; n-damo细菌丰度较高,16S rRNA基因拷贝数可达1.25~2.31×10~8拷贝每克干重,pmo A基因拷贝数为8.66×10~6~2.54×10~7拷贝每克干重。研究为后续富集培养嗜酸型n-damo菌提供可靠菌源。

【Abstract】 The recently discovered nitrite-dependent anaerobic methane oxidizer( n-damo) has a promising application capability in the field of nitrogen removal of wastewater. This study focuses on detecting the diversity and abundance of n-damo bacteria in acidic paddy soil,using phylogenetic analysis and quantitative polymerase chain reaction technology based on n-damo-specific 16 S rRNA gene and pmoA gene. The results show that n-damo bacteria clusters into 3-4 OTUs in the paddy soil samples with weak acidity and moderate nutrient level. Phylogenetic analysis indicates that novel unknown n-damo bacteria exist in the acidic paddy soil. The abundance of n-damo bacteria is generally high,with 16 S rRNA gene and pmoA gene in the range of1. 25 ~ 2. 31 × 10~8 and 8. 66 × 10~6~ 2. 54 × 10~7 copies per g dry weight,respectively. Our study offers reliable source of inoculum for subsequent enrichment of acidophilic n-damo bacteria.
