长链非编码RNA ATB检测在乳腺癌诊断中的意义
Value of detection of long non-coding RNA ATB in the diagnosis of breast cancer
【摘要】 目的探讨长链非编码RNA(1ncRNA) ATB在乳腺癌患者血清中的水平及其诊断乳腺癌的潜在价值。方法采用实时荧光定量PCR(q PCR)检测2017年6月至2019年9月37例乳腺癌患者、30例乳腺良性疾病患者和26例健康体检者血清1ncRNA ATB的水平,分析1ncRNA ATB表达量与乳腺癌临床病理特征的关系,采用受试者工作特征(ROC)曲线分析1ncRNA ATB对乳腺癌的诊断价值。结果血清1ncRNA ATB在乳腺癌组[1.82(1.19,3.74)]、乳腺良性疾病组[1.13(0.92,1.31)]和健康体检组[0.98(0.83,1.14)]中水平的差异有统计学意义(P<0.01)。乳腺癌患者血清1ncRNA ATB的表达水平与ER、c-erbB-2、HER-2、TNM分期和淋巴结转移有关(P<0.05),与年龄、肿瘤大小和PR无关(P<0.05)。单独检测1ncRNA ATB的曲线下面积(AUC)为0.824 (95%CI:0.735~0.914),灵敏度为89.2%,特异度为67.9%; 1ncRNA ATB联合CA153检测的AUC为0.876(95%CI:0.800~0.953),灵敏度为83.8%,特异度为80.4%。26例乳腺癌患者术后1周1ncRNA ATB水平为[1.36(1.09,1.59)],较术前[1.93(1.27,3.94)]明显降低,差异有统计学意义(P<0.01)。结论乳腺癌患者血清1ncRNA ATB高表达,有望成为乳腺癌诊断的生物标志物。
【Abstract】 Objective To investigate the level of serum long non-coding RNA(lncRNA) ATB in patients with breast cancer and its potential value for the diagnosis of breast cancer. Methods The levels of lncRNA ATB were determined by quantitative reverse real-time PCR(q PCR) in the serum of 37 patients with breast cancer,30 patients with benign breast diseases and 26 healthy controls.The correlation between serum 1 ncRNA ATB levels and clinicopathological characteristics of breast cancer was analyzed. Receiver operating characteristic(ROC) curve was used to analyze the diagnosis value of lncRNA ATB in breast cancer. Results The relative expression level of serum 1 ncRNA ATB in breast cancer patients was [1. 82(1. 19,3. 74) ],higher than [1. 13(0. 92,1. 31) ] in benign breast diseases patients and [0. 98(0. 83,1. 14) ]in healthy controls(P<0. 01). Moreover,the level of serum 1 ncRNA ATB in breast cancer patients was correlated with the expression of ER,c-erbB-2,HER-2,TNM staging and lymph node metastasis(P<0. 05),but not with age,tumor size and PR. The area under curve(AUC) of serum 1 ncRNA ATB for the diagnosis of breast cancer was 0. 824(95%CI: 0. 735-0. 914) with sensitivity of 89.2% and specificity of 67.9%. The AUC of serum 1 ncRNA ATB in combination with CA153 for breast cancer detection was 0. 876(95%CI: 0. 800-0. 953) with sensitivity of 83. 8% and specificity of 80. 4%. Among26 patients with breast cancer,the post-operative serum level of 1 ncRNA ATB was [1. 36(1. 09,1. 59) ],lower than [1. 93(1. 27,3. 94) ] before operation(P<0. 01). Conclusion The level of serum 1 ncRNA ATB in breast cancer patients increases,which may service as a potential biomarker for the diagnosis of breast cancer.
