【摘要】 [目的]探讨5-氮杂-2′-脱氧胞苷(5-Aza-CdR)对胃癌SGC-7901细胞增殖的抑制作用及ERCC1 mRNA的影响。[方法]采用实时荧光定量RT-PCR分别检测使用5-Aza-CdR处理胃癌SGC-7901细胞前后细胞内ERCC1 mRNA表达水平。MTT法检测单独使用5-Aza-CdR或顺铂及两药联合使用对胃癌SGC-7901细胞的抑制作用。[结果]不同浓度的5-Aza-CdR对SGC-7901人胃癌细胞的生长均有抑制作用,抑制率与对照组相比,差异有统计学意义(P<0.05);5-Aza-CdR也显著抑制SGC-7901人胃癌细胞ERCC1 mRNA的表达,其抑制作用呈时间-剂量依赖;顺铂联合5-Aza-CdR可使SGC-7901人胃癌细胞对顺铂的IC50值下降。[结论]5-Aza-CdR联合顺铂能协同性地抑制胃癌细胞生长,这可能与5-Aza-CdR能影响ERCC-1 mRNA的表达有关。

【Abstract】 [Objective]To explore the inhibitory effects of 5-Aza-CdR on the proliferation as well as ERCC1 expression of gastric cancer SGC7901 cells.[Methods]Real-time fluorescent quantitative PCR was used to detect the expression of excision repair cross complementary gene 1(ERCC1)mRNA of in different groups.MTT assay was used to assess the inhibitory effects of 5-Aza-CdR,cisplatin alone or a combination of both on gastric cancer SGC7901 cells.[Results]5-Aza-CdR significantly inhibited the growth of SGC-7901 human gastric cancer cells.It elevated the inhibiting rate(P< 0.05)as compared with control groups.The expression of ERCC1 mRNA was significantly inhibited in SGC-7901 human gastric cancer cells and xenograft in dose and time dependent manner.Cisplatin combined with 5-Aza-CdR elevated significantly the IC50 of SGC-7901 human gastric cancer cells as compared with cisplatin alone.[Conclusions]The investigation suggests that a synergrstic offecto of combination of 5-Aza-CdR and cisplatin induced cell growth inhibition,which may be resulted from the expression of ERCC1 influencing by 5-Aza-CdR
