两种培养基在双层溶血环实验中的应用效果比较
Comparison of two kinds of media in the experiment of double hemolytic ring
【摘要】 目的在双层溶血环实验中获得清晰、亮丽的双层溶血环效果,并能稳定地维持较长时间,以便能更好地示教给学生用,及减少医学微生物学实验教学中的准备工作量。方法用改良厌氧罐培养,将同一株产气荚膜杆菌接种于苯乙醇厌氧培养基(EPA)与自制的哥伦比亚兔血琼脂平板(CRBA),共罐同等厌氧条件下培养,18~24 h后取出,进行为期1周或更长时间的效果比较。结果 EPA培养出的双层溶血环维持时间更长、更稳定,环也很亮丽、透明,界限分明。结论苯乙醇厌氧培养基在双层溶血环实验教学中的应用效果更简易、可行、符合教学要求及3R规则的。
【Abstract】 Objective To obtain a clear and bright double-layer hemolysis loop effect in the experiment of double-layer hemolysis loop,and to maintain a stable effect for a long time,so as to show it to students better,and to reduce the workload of preparation in the experiment teaching of medical microbiology.Methods The same strain of perfringens was inoculated into phenylethanol anaerobic medium (EPA) and self-made Columbia rabbit blood agar plate (CRBA) and cultured in a total tank under the same anaerobic condition.After 18-24h,it was taken out to observe the double hemolytic loop for a period of one week or longer time.Results The double-layer hemolytic loop with bright,transparent and well-defined,maintained longer and more stable.Conclusion The application effect of phenylethanol anaerobic medium in the teaching of double-layer hemolytic loop experiment,is more simple,feasible,in line with teaching requirements and 3R rules.
