主动脉夹层病人PDLIM3基因的表达差异及临床意义

【摘要】 目的探讨主动脉夹层(aortic dissection,AD)病人主动脉壁中PDLIM3的表达差异及其临床意义。方法 2017年1月~2018年12月我院收治主动脉夹层病人34例,心脏移植受体主动脉组织标本22例,组织切片经HE染色及弹性纤维染色(EVG)评价主动脉壁结构改变;利用实时荧光定量PCR(RT-PCR)检测主动脉样本中PDLIM3基因的mRNA表达水平,并进一步分析PDLIM3的表达量与主动脉夹层病人主动脉直径的相关关系。结果 HE及EVG染色可见主动脉夹层病人主动脉壁中层变薄,弹性纤维断裂增加,排列紊乱,血管平滑肌细胞数目减少,体积变小,形态改变;与移植受体比较,主动脉夹层病人主动脉壁中PDLIM3基因的mRNA表达水平显著下调(P<0.05);Pearson相关性分析发现,PDLIM3基因表达与病人主动脉直径无相关性。结论主动脉夹层病人主动脉壁中PDLIM3基因表达下调,可能与主动脉夹层的发生有关。

【Abstract】 Objective To investigate the expression pattern of PDLIM3 in the aorta of patients with aortic dissection(AD) and its clinical significance.Methods Aortic tissue specimens from aortic dissection patients(n=34) and heart transplant recipients(n=22) who received surgical treatment in Tongji Hospital from January 2017 to December 2018 were collected.The structural changes of aortic wall were observed by hematoxylin-eosin(HE) staining and elastic fiber staining(EVG).Real-time quantitative PCR(RT-PCR) was used to detect the mRNA expression levels of PDLIM3 in aortic samples.The correlation between the expression levels of PDLIM3 and the aortic diameter was analyzed by using Pearson correlation analysis.Results HE and EVG staining showed that the middle layer of aortic wall became thinner, elastic fiber fracture increased, vascular smooth muscle cellloss and with smaller volume and morphology changes.The mRNA expression levels of PDLIM3 in aorta of patients with aortic dissection were significantly down-regulated compared with that of the transplanted recipients(P<0.05).Pearson correlation analysis showed that the expression of PDLIM3 gene was not significantly associated with the aortic diameter of patient with aortic dissection.Conclusion PDLIM3 expression level is down-regulated in aorta of patients with aortic dissection and may be associated with the occurrence of aortic dissection in patients.
