纳米混悬剂中羟基喜树碱和槲皮素的含量测定及方法学考察
Determination and methodological study of hydroxycamptothecin and quercetin in nano-suspension
【摘要】 [目的]使用高效液相色谱法,建立纳米混悬剂中羟基喜树碱和槲皮素的含量测定方法。[方法]采用SinoChrom ODS-BP色谱柱(250 mm×4.6 mm,5μm);流动相为甲醇-水(60∶40);检测波长为374 nm,柱温30℃;流速1.0 mL·min-1;进样量20μL。并对该方法的精密度、回收率、加样回收率进行考察。羟基喜树碱的线性回归方程为:Y=81 268 X-10 761(R2=0.999 9),线性范围为0.506~10.12μg·mL-1;槲皮素的线性回归方程为:Y=67 343 X-8 960.3(R2=0.999 9),线性范围为0.51~10.2μg·mL-1。[结果]羟基喜树碱和槲皮素在线性范围内色谱峰面积与浓度均有很好的线性关系。并在保留时间内未见其他干扰峰,该方法的适应性、精密度和回收率均较好。[结论]该方法可以用于同时测定羟基喜树碱和槲皮素的含量。
【Abstract】 [Objective] To establish a determination method of hydroxycamptothecin and quercetin in nano-suspension by HPLC. [Methods]the SinoChrom ODS-BP column(250 mm×4.6 mm, 5 μ m) was used, the mobile phase was methanol-water(60∶40), the detection wavelength was 374 nm, the column temperature was 30 ℃, the flow rate was 1.0 mL/min, and the injection volume was 20 μL. The precision, recovery and sample recovery of the method were investigated. [Results] the linear regression equation of hydroxycamptothecin was Y = 81 268 X-10 761(R2 = 0.999 9). The linear regression equation of quercetin was 0.506~10.12 μg·mL-1;. The linear regression equation of quercetin was Y = 67 343 X-8 960.3(R2 = 0.999 9), and the linear range was 0.51~10.2 μg·mL-1. The results showed that there was a good linear relationship between the chromatographic peak area and the concentration of hydroxycamptothecin and quercetin in the linear range. There are no other interference peaks in the retention time, and the adaptability, precision and recovery of the method are good.[Conclusion] this method can be used for the simultaneous determination of hydroxycamptothecin and quercetin.
