五味子醇甲对转tau基因果蝇阿尔兹海默病模型dTORmRNA表达的影响

【摘要】 目的研究五味子醇甲对转tau基因果蝇dTORmRNA表达的影响。方法转tau果蝇分为模型组和五味子组,正常果蝇为空白组,给予对应的培养基进行饲养,7 d后对果蝇进行嗅觉学习记忆能力测试,检测相关指标表达情况。结果模型组学习记忆能力最低,其余两组能力相仿;空白组dTORmRNA表达水平最高,五味子组次之,模型组最低;五味子组果蝇dS6K和d4E-BP表达水平低于模型组。结论五味子醇甲可有效调节转tau基因果蝇dTORmRNA表达水平,降低dS6K和d4E-BP蛋白表达,提高转tau基因果蝇嗅觉学习记忆能力,改善转tau基因果蝇阿尔兹海默病症状。

【Abstract】 Objective To study and analyze the effect of schisandrin on the expression of dTORmRNA in drosophila model of Alzheimer’s disease.Methods Transgenic fruit flies are divided into model group and Schisandra group.Normal fruit flies are the blank group and are fed with the corresponding medium.After 7 d,the fruit flies are tested for olfactory learning and memory ability to detect the expression of related indicators.Results The model group has the lowest learning and memory ability,and the other two groups have similar abilities;the blank group has the highest dTORmRNA expression level,followed by the schisandrae group,and the model group has the lowest;the dS6K and d4E-BP expression levels of fruit flies in the schisandra group are lower than the model group.Conclusion Schisandrin can effectively regulate the expression level of dTORmRNA in transgenic flies with tau gene,reduce the expression of dS6K and d4E-BP protein,improve the olfactory learning and memory ability of transgenic flies with tau gene,and improve the symptoms of Alzheimer’s disease in transgenic flies with tau gene.
