白藜芦醇诱导白念珠菌凋亡的机制探讨
Mechanism of resveratrol induced apoptosis of Candida albicans
【摘要】 目的探究白藜芦醇诱导白念珠菌凋亡的机制。方法将不同浓度的白藜芦醇(终浓度0.256、0.128、0.064mg/mL)和作为阳性对照的两性霉素B(AmB,终浓度0.5μg/mL)分别加入指数生长期的白念珠菌悬液,同时设置空白对照组,检测白念珠菌的凋亡率、细胞内活性氧和线粒体膜电位等指标。结果与空白对照组比较,白藜芦醇处理后的白念珠菌凋亡率、活性氧水平明显升高,线粒体膜电位明显降低,差异均有统计学意义(P<0.05)。结论白藜芦醇具有诱导白念珠菌凋亡的作用,这可能与活性氧积累和线粒体损伤相关。
【Abstract】 Objective To explore the mechanism of resveratrol induced apoptosis of Candida albicans.Methods Different concentrations of resveratrol(final concentrations were 0.256,0.128,0.064 mg/mL)and amphotericin B(AmB,final concentration was 0.5μg/mL)which was used as the positive control were added into the bacterial suspension at exponential growth stage respectively,meanwhile,the blank group was set up.Then the apoptosis rate of Candida albicans,intracellular reactive oxygen species and mitochondrial membrane potential were measured.Results Compared with the blank group,the apoptotic rate of Candidaalbi canstreated with resveratrol,the level of intracellular ROS increased significantly,and the mitochondrial membrane potential decreased significantly(P<0.05).Conclusion Resveratrol can induce the apoptosis of Candida albicans,which may be related to the accumulation of ROS and mitochondrial damage.
