芘对斑马鱼胚胎毒性的研究
Effect of Pyrene on the Expression of Vitellogenin of Larva Zebrafish
【摘要】 以斑马鱼的卵黄蛋白原(Vtg)为生物标志物,研究芘对斑马鱼胚胎毒性.用100、200和300μg·L-1 3种不同浓度的芘和无水乙醇(空白对照)同时处理受精9 h后的斑马鱼胚胎,暴露至72 h与144 h,观察并统计斑马鱼胚胎孵化率、死亡率以及畸形率数据以及Vtg基因表达的情况.实验结果表明:芘可以影响斑马鱼胚胎的发育,芘的暴露导致斑马鱼胚胎出现心包水肿、卵黄囊水肿、脊柱弯曲以及尾部弯曲等中毒症状.暴露72 h和144 h后,随着芘浓度升高斑马鱼胚胎孵化率下降,畸形率以及死亡率上升,而且中毒程度随暴露时间的延长而增加.与对照相比,芘对斑马鱼Vtg基因表达量均下降,Vtg基因均没有被显著诱导.
【Abstract】 Vitellogenin(VTG) of zebrafish was used as a biomarker to study the toxicity of pyrene on zebrafish embryo. Zebrafish embryos after fertilization for 9 h were treated with 100, 200 and 300 μg·L-1 pyrene and anhydrous ethanol(blank control) at the same time. When exposed for 72 h and 144 h, the hatchability, mortality, deformity rate and VTG gene expression of zebrafish embryos were observed and counted. The results showed that pyrene could affect the development of zebrafish embryos, and pyrene exposure led to pericardial edema, yolk sac edema, spinal curvature, tail curvature and other toxic symptoms in zebrafish embryos. After exposure for 72 h and 144 h, with the increase of pyrene concentration, the hatching rate of zebrafish embryos decreased, the deformity rate and mortality rate increased, and the degree of poisoning increased with the extension of exposure time; compared with the control subject, the expression of VTG gene in zebrafish decreased, but VTG gene was not significantly induced.
