电热损伤法建立新西兰大白兔宫腔内膜损伤模型

【摘要】 目的建立一个稳定有效的宫腔内膜损伤动物模型,为研究宫腔内膜损伤的发病机理及治疗方法奠定基础。方法75只成年雌性新西兰大白兔随机分为3组:电热损伤组25只、机械损伤组25只、对照组25只。机械损伤组刮宫损伤子宫内膜,电热损伤组采用电刀损伤子宫内膜,对照组行假手术。为评估建模效果,分别在术后7d(每组5只),14d(每组5只),28d(每组5只)比较各组的子宫内膜腺体数量、纤维化面积,于术后7d配种,一部分(每组5只)配种后14d取子宫比较各组着床胚胎数,一部分(每组5只)饲养至分娩观察分娩仔兔数。结果术后7d和14d,与机械损伤组和对照组比较,电热损伤组子宫内膜腺体数量显著减少(P<0.05),内膜纤维化面积显著增大(P<0.05),差异有统计学意义;术后28d 3组比较子宫内膜腺体数量及子宫内膜纤维化面积,差异无统计学意义(P>0.05)。与机械损伤组和对照组比较,电热损伤组着床胚胎数更少,分娩的仔兔更少,差异有统计学意义(P<0.05)。结论电热损伤后7-14d造成的兔宫腔内膜损伤模型稳定有效。

【Abstract】 Objective The pathogenesis and treatment of intrauterine injury remain unsolved, highlighting the need for stable and effective experimental animal models. Methods In this study, 75 female New Zealand White rabbits were randomly divided into electrothermal group,(n=25), mechanical injury group(n=25) and the control group(n=25). The number of endometrial glands,area of endometrial fibrosis, number of implanted fetuses and number of birth were compared among the 3 groups on day 7(5 rabbits/group), day 14(5 rabbits/group) and day 28(5 rabbits/group) after surgery. Results In the electrothermal group, the numbers of endometrial glands on Days 7 and 14 and the number of implanted fetuses and births were significantly lower than those in mechanical injury group and the control group(P<0.05). While compared with the mechanical injury group and the control group, in the electrothermal group, the ratio of the area with endometrial stromal fibrosis to the total endometrial area on Days 7 and 14 was significantly increased(P <0.05). And on Days 28, the number of endometrial glands, area of endometrial fibrosis among the 3 groups were not significantly different(P>0.05). Conclusion These results suggest that electrothermal injury is effective for the establishment of a rabbit intrauterine injury model between 7 and 14 d after surgery.
