miRNA-19基因表达与大脑中动脉M1段狭窄程度及斑块易损性的相关性
Relationship between miRNA-19 Gene Expression and M1 Stenosis Degree and Plaque Vulnerability in Middle Cerebral Artery
【摘要】 目的分析miRNA-19a、miRNA-19b-1与大脑中动脉M1段狭窄程度及斑块易损性的相关性。方法选取大脑中动脉M1段狭窄患者(病例组)和正常者(对照组)各80例;根据动脉狭窄程度将患者分为轻度组、中度组和重度组;根据斑块易损性将患者分为高危组、中危组和低危组。采用qPCR检测各组血清miRNA-19a、miRNA-19b-1的表达水平,受试者工作特征曲线分析二者预测高危斑块的价值,计算曲线下面积(AUC)。结果血清miRNA-19a和miRNA-19b-1水平比较:病例组高于对照组;重度组高于轻度和中度组,中度组高于轻度组;高危组高于低危组和中危组,中危组高于低危组(均P<0.05)。miRNA-19a和miRNA-19b-1表达与动脉狭窄程度(r=0.826、0.795,P<0.05)、斑块易损危险性(r=0.701、0.658,P<0.05)呈正相关。miRNA-19a和miRNA-19b-1预测高危斑块的AUC分别为0.774和0.685,二者联合检测可将AUC提高至0.814。结论血清miRNA-19a、miRNA-19b-1与大脑中动脉M1段狭窄程度及斑块易损性密切相关,提示血清miRNA-19a、miRNA-19b-1可能是动脉狭窄和斑块易损的影响因素,可用于预测大脑中动脉M1段高危斑块。
【Abstract】 Objective To explore the correlation between expression of miRNA-19 a and miRNA-19 b-1 and M1 stenosis degree and plaque vulnerability in middle cerebral artery.MethodsEighty patients with M1 stenosis of middle cerebral artery were selected as the case group,and 80 normal subjects as the control group.According to the degree of arterial stenosis,patients were divided into mild group,moderate group and severe group.According to plaque vulnerability,patients were divided into high-risk group,medium-risk group and low-risk group.The expression of miRNA-19 a and miRNA-19 b-1 was detected by qPCR.The receiver operating characteristic curve was used to analyze the values of miRNA-19 a and miRNA-19 b-1 in predicting high-risk plaques,and the area under the curve(AUC) was calculated.Results Serum levels of miRNA-19 a and miRNA-19 b-1 in case group were higher than those in control group,those in severe group were higher than those in mild or moderate group,and those in moderate group were higher than those in mild group(P<0.05).In addition,miRNA-19 a and miRNA-19 b-1 levels in high-risk group were higher than those in low-risk or medium-risk group,and those in medium-risk group were higher than those in low-risk group(P<0.05).Serum miRNA-19 a and miRNA-19 b-1 expression was positively correlated with arterial stenosis degree(r=0.826 and 0.795,respectively; P<0.05) and plaque vulnerability risk(r=0.701 and 0.658,respectively;P<0.05);The AUCs of miRNA-19 a,miRNA-19 b-1 and their combination in predicting high-risk plaque were 0.774,0.685 and 0.814,respectively.Conclusion Serum miRNA-19 a and miRNA-19 b-1 levels are closely related to plaque vulnerability and M1 stenosis of middle cerebral artery,suggesting that serum miRNA-19 a and miRNA-19 b-1 may be the influencing factors for plaque vulnerability and M1 stenosis and can be used to predict high-risk plaque in M1 segment of middle cerebral artery.
