一株沙门菌裂解性噬菌体的分离鉴定与生物学特性研究
Isolation, Identification and Biological Characteristics of A Salmonella Lytic Phage
【摘要】 以1株多重耐药沙门菌(SA209)为宿主菌,从安阳市某规模化养殖场污水样本中分离出1株沙门菌裂解性噬菌体,命名为SA-1。采用点滴法和双平板法分离、鉴定及纯化了SA-1,并对其效价、最佳感染复数、酸碱稳定性、热稳定性、一步生长曲线等生物学特性进行了研究。结果表明:SA-1的初始效价为1.15×1010 PFU/mL;最佳感染复数为0.001;对酸碱的耐受性较强,在pH 3.0~11.0范围内活性稳定;在40~50℃温度内活性稳定;其感染宿主菌的潜伏期为10 min,裂解期为20 min,之后进入稳定期,平均裂解量为108 PFU/cell; SA-1的核酸类型为DNA。
【Abstract】 A multi-drug resistant Salmonella(SA209) was used as the host bacteria to isolate a strong Salmonella bacteriophage from the sewage of a large-scale farm in Anyang City, named SA-1. The phage SA-1 was separated, identified and purified by spot method and double-plate method, and the biological characteristics of the bacteriophage SA-1 titer, optimal multiplicity of infection, acid-base stability, thermal stability, and one-step growth curve were studied. The results showed that the initial titer of SA-1 was 1.15×1010 PFU/mL. The optimal multiplicity of infection was 0.001. It had a wide tolerance to acids and bases y in the range of pH 3.0~11.0, and had stable internal activity at a temperature of 40 ~50 ℃. The result of the one-step growth curve showed that the incubation period of the infected host bacteria was 10 minutes. The lysis period was 20 minutes, and then entered the stable period. The average lysis amount was 108 PFU/cell. SA-1 nucleic acid type was DNA.
