小麦TaNAC-B072基因的克隆和表达分析

【摘要】 为了研究小麦NAC转录因子的功能及其对逆境胁迫的响应,本试验克隆了小麦的TaNAC-B072基因。该基因的开放阅读框长度为978 bp,编码蛋白长度为325个氨基酸,推测其分子量为35.75 kDa,等电点为7.06,属于中性蛋白,具有1个NAC结构域。多序列比对和系统进化树分析显示,TaNAC-B072与BdNAC72-like的亲缘关系较近。RT-PCR结果显示,在小麦的根和地上部分,TaNAC-B072基因的表达受高盐、渗透和冷胁迫的诱导,初步推测基因TaNAC-B072可能与小麦响应逆境胁迫有密切的关系。

【Abstract】 In order to study the function of wheat NAC gene and it response to stress, the TaNAC-B072 gene of wheat was cloned in the experiment. Sequence analysis showed that the open reading frame(ORF) of TaNAC-B072 was 978 bp, encoding 325 amino acids. Its protein contained a typical NAC domain, with molecular weight of 35.75 kDa and isoelectric point of 7.06. Multiple sequence alignment and phylogenetic tree analysis showed that TaNAC-B072 was closely related to the BdNAC72-like gene. The RT-PCR analysis showed that the expression of TaNAC-B072 was induced or inhibited by NaCl, osmotic and cold stress in root and shoot tissue, which suggested that the TaNAC-B072 gene might be closely related to plant resistance to abiotic stress.
