秦巴山区魔芋白绢病病原菌的鉴定及生物学特性的测定
Identification and Biological Characteristics of Hypochnus centrifugus in Qinba Mountains Area
【摘要】 对秦巴山区魔芋白绢病病原菌进行了分离、鉴定和生物学特性测定。结果表明,菌株M3具有致病性,菌落正面白色,菌丝发达呈放射状,背面呈淡浅黄色,后期形成茶褐色菜籽状菌核,直径为1.2~2.3 mm,具有蘑菇气味;经16 S rDNA-ITS序列相似性分析,该病原菌与罗耳阿太菌Athelia rolfsii(KY640625.1)相似性达100%,结合形态学特征,将其鉴定为罗耳阿太菌Athelia rolfsii,属于半知菌亚门,其无性态为齐整小核菌Sclerotium rolfsii;菌株M3的最适生长温度为28℃,最适生长pH值为7,碳源利用以麦芽糖最好,乳糖最差;氮源利用以酵母粉最好,硝酸铵最差。该研究结果为秦巴山区魔芋白绢病的绿色防控和综合治理提供了依据。
【Abstract】 The pathogen of Hypochnus centrifugus in Qinba Mountains area was isolated and identified, and its biological characteristics was also determined. The results showed that the strain M3 was pathogenic. The colony was white on the front, and the hyphae were radiate. The back was light yellow, and sclerotia were brown in the later stage, diameter were 1.2~2.3 mm. It had mushroom odor. The 16 S rDNA-ITS sequence of strain M3 shared similarity was 100% with Athelia rolfsii(KY640625.1) that had reported. So, the pathogen was identified as Athelia rolfsii combined the morphologic characteristics with ITS sequence analysis. The optimal temperature for strain M3 was 28 ℃. The optimal pH value was 7. The best carbon source and nitrogen source were maltose and yeast powder, respectively. The worst were lactose and ammonium nitrate. The results provide a basis for the diagnosis and integrated control of Hypochnus centrifugus in Qinba Mountains area.
