云南糯玉米地方品种糯性等位基因wx-xuanwei的分子特征

【摘要】 为了解云南糯玉米地方品种糯性waxy基因的等位变异类型与构成,有针对性地保护和利用携带稀有基因的珍稀遗传材料,在用糯玉米已知基因wx-D7、wx-D10、wx-Cin4、wx-124和wx-Reina等的特异分子标记检测并获得携带有未知糯性基因的云南糯玉米地方品种基础上,设计出能检测waxy全基因的11对分子标记,检测10份带有未知waxy等位基因的云南糯玉米地方品种,发现来自云南曲靖市辖宣威市的糯玉米地方品种糯包谷(统一编号232248)带有的waxy基因的突变位点。用简化的热交错不对称PCR(Tail-PCR)对该突变位点作进一步扩增和测序,并分析包含突变点在内的waxy基因的分子特征。宣威糯包谷的waxy基因在其第14外显子中插入了约4.9 kb的未命名反转录转座子,本研究将其命名为xuanwei(宣威)。该反转录转座子属于长末端重复(Long terminal repeat,LTR)反转录转座子,具有序列完全相同的LTR结构。因此waxy基因wx-xuanwei是一个反转录转座子插入造成的新的等位突变基因。

【Abstract】 In order to understand the allele diversity and composition of waxy alleles in waxy maize landraces in Yunnan Province, the precious genetic materials with rare alleles should be protected and utilized. Based on identification of Yunnan waxy maize landraces with unknown waxy genes by the detection of specific molecular markers of known waxy maize genes including wx-D7, wx-D10, wx-Cin4, wx-124 and wx-Reina, 10 Yunnan waxy maize landraces with unknown waxy alleles were detected and used in this study. 11 pairs of molecular markers were designed to detect the whole waxy gene and the waxy allele mutation of Nuobaogu(accession number is 232248) from Xuanwei County, Qujing City, Yunnan Province was found. Simplified thermal asymmetric interlaced PCR(tail-PCR) was used to amplify the mutation, and the molecular characteristics of the waxy gene mutation was analyzed by sequencing. There was an about 4.9 kb unnamed retrotransposon insertion in exon 14 of waxy gene in xuanwei Nuobaogu, and this retrotransposon was named xuanwei in this study. The retrotransposon belongs to the long terminal repeat(LTR) retrotransposon and has the same LTR sequence. Therefore, the waxy gene wx-xuanwei is a new allele mutation caused by the insertion of retrotransposons.
