HPLC法同时测定蒿茶中3种有机酸的含量

【摘要】 建立蒿茶中新绿原酸、绿原酸、隐绿原酸3种有机酸的HPLC含量测定方法。采用高效液相色谱法,色谱柱:Kromasil 100-5C18柱(250 mm×4. 6 mm,5μm);流动相:乙腈-0.1%甲酸水(10︰90,V/V);流速:1.0 m L·min-1;检测波长:326nm;柱温:30℃。试验结果表明,3种有机酸的进样量与峰面积呈良好的线性关系,平均加样回收率在96. 9%~104. 6%,RSD在2. 0%~2. 4%。不同产地的样本中,江苏蒿茶的3种有机酸类成分的含量远大于其他4个产地。本方法简便易行、准确可靠,可用于蒿茶中3种有机酸的含量测定。

【Abstract】 The High performance liquid chromatography( HPLC) method for determination of three kinds of organic acids,namely,neochlorogenic acid,chlorogenic acid and cryptochlorogenic acid in Artemisia tea was established. The conditions of HPLC method were as follows: the chromatographic column of Kromasil 100-5 C18( 250 mm × 4. 6 mm,5 μm),the mobile phase of acetonitrile-0. 1% formic acid water( 10 ︰ 90,V/V) at the rate of 1. 0 m L·min-1,the detection wavelength of 326 nm and the column temperature of 30℃ . The results showed that there was a good linear relationship between the injection volume of three organic acids and the peak area. The average recovery rate was between 9 6. 9 % and 1 0 4. 6 %,and the RSD was between 2. 0 % and2. 4%. Among the samples from different producing areas,the contents of three organic acids in Artemisia tea from Jiangsu were much higher than those from the other four producing areas. Conclusions: The established method of HPLC is simple,accurate and reliable to determine the content of organic acids in Artemisia tea.
