石蒜碱对人肺癌NCI-H460细胞增殖和凋亡的影响
Effects of lycorine on proliferation and apoptosis of human lung cancer NCI-H460 cells
【摘要】 目的探讨石蒜碱对人肺癌NCI-H460细胞增殖和凋亡的影响。方法人肺癌NCI-H460细胞株于DMEM培养基进行培养,采用1.25、2.5、5.0、10.0、20.0μmol/L的石蒜碱对其进行处理,并以0.9%NaCl溶液为对照组。倒置显微镜下观察NCI-H460细胞形态变化,MTT法检测细胞增殖活性,流式细胞仪检测细胞凋亡率,Western blot法检测细胞凋亡相关基因p53、Bcl-2、Bax、Caspase-2、Caspase-3的蛋白表达水平。结果经石蒜碱处理后,NCI-H460细胞外形不规则、细胞皱缩、大小不一致,且细胞破碎程度随给药剂量增加而上升,细胞数量有减少趋势。石蒜碱处理24 h、48 h后,NCI-H460细胞增殖活性显著降低,且下降趋势呈剂量与时间依赖性。石蒜碱处理24 h后,NCI-H460细胞凋亡率显著增高,NCI-H460细胞p53、Bax、Caspase-2、Caspase-3蛋白表达水平显著上调,而Bcl-2表达水平显著下调。结论石蒜碱抑制NCI-H460细胞增殖,促进凋亡,且呈时间与剂量依赖性,与调控凋亡相关蛋白有关。
【Abstract】 Objective To study the effects of lycorine inhibition on proliferation and apoptosis of human lung cancer NCI-H460 cells. Methods Human lung cancer NCI-H460 cell lines were cultured in DMEM medium. The cells were treated with 1.25, 2.5, 5.0, 10.0, 20.0 μmol/L lycorine and 0.9% NaCl solution as control group. The morphological changes of NCI-H460 cells were observed under inverted microscope. MTT analysis was used to detect cell proliferation activity, flow cytometry was used to detect apoptosis rate, Western blot was used to detect the protein expression of p53, Bcl-2, Bax, caspase-2 and caspase-3. Results After treatment with lycorine, NCI-H460 cells were irregular in shape, shrunk and inconsistent in size, and the degree of cell fragmentation was increased in dose dependent manner, and the number of cells was decreased. The proliferation activity of NCI-H460 cells was decreased significantly 24 h and 48 h after treatment with lycorine, and the decreasing trend was dose-dependent and time-dependent(P<0.05). 24 hours after lycorine treatment, the apoptosis rate of NCI-H460 cells was increased significantly, the expression levels of p53, Bax, caspase-2 and caspase-3 proteins in NCI-H460 cells were increased significantly, but decreased for Bcl-2(P<0.05). Conclusion Lycorine inhibited the proliferation and promoted apoptosis of NCI-H460 cells, which is related to the regulation of apoptosis related proteins.
