MTHFR C677T、MTHFR A1298C、MTRR A66G位点基因多态性与H型高血压患者Hcy、血脂水平的关系
Association of MTHFR C677T, MTHFR A1298C, MTRR A66G site gene polymorphism with Hcy and lipids in patients with H-type hypertension
【摘要】 目的探讨亚甲基四氢叶酸还原酶(MTHFR)C677T、A1298C,5-甲基四氢叶酸高半胱氨酸甲基转移酶还原酶(MTRR)A66G位点基因多态性与H型高血压患者Hcy、血脂水平的关系。方法入选2018年9月~2019年7月140例以高血压病收治入院的患者,收集一般临床资料和血生化检查结果,高效液相色谱荧光法测定同型半胱氨酸(Hcy)水平,其中82例为H型高血压患者。将这82例患者PCR产物进行基因测序,以PCR-芯片杂交法测定其MTHFR基因多态性。观察MTHFR C677T、MTHFR A1298C、MTRR A66G位点不同基因型的Hcy、血脂水平的差异。结果 MTHFR C677T中T/T基因型的血浆Hcy水平与C/C、C/T基因型比较的差异有显著性意义(P<0.01),MTHFR C677T中T/T基因型血浆TC、TG、HDL及LDL无明显差异(P>0.05);MTHFR A1298C、MTRR A66G的三种基因型患者的Hcy、TC、TG、HDL及LDL水平差异无统计学意义(P>0.05)。结论 H型高血压患者中MTHFR C677T位点基因多态性与血脂异常无关联,而与Hcy呈显著相关,MTHFR A1298C、MTRR A66G位点基因多态性与Hcy和血脂均无相关性。
【Abstract】 Objective To investigate the relationship between Hcy and lipids with the polymorphism of MTHFR C677 T, MTHFR A1298 C, MTRR A66 G in the patients with H-type hypertension. Methods A total of 82 of 140 patients with H-type hypertension were selected as the research subjects from September, 2018 to July, 2019. The personal information was collected, clinical biochemical indicators were tested,including plasma Hcy, lipids. The Hcy was determined by HPLC. The gene polymorphism of MTHFR C677 T, MTHFR A1298 C, MTRR A66 G site was detected by PCR-chip hybridization. The Hcy and lipids levels with gene polymorphism of MTHFR and MTRR were observed. Results The concentrations of Hcy in the patients with T/T genotype of MTHFR C677 T were significantly higher than those with C/C and C/T genotypes of MTHFR C677 T, but those of Hcy showed no significant difference in MTHFR A1298 C, MTRR A66 G genotypes. The concentrations of TC, TG, HDL and LDL showed no significant difference in the patients with T/T, C/C and C/T genotypes of MTHFR C677 T, the same as those genotypes of MTHFR A1298 C, MTRR A66 G. Conclusion Polymorphism of MTHFR C677 T is closely related to plasma Hcy levels, isn’t associated with lipids. Neither Hcy or lipids is related to the polymorphism of MTHFR A1298 C, MTRR A66 G.
