血清miR-21和miR-125b预测乳腺癌新辅助化疗疗效和预后
Serum miR-21 and miR-125b predict neoadjuvant chemotherapy response and prognosis in breast cancer
【摘要】 目的研究血清miRNA对乳腺癌新辅助化疗反应和预后的预测价值。方法本研究纳入102例接受新辅助化疗的乳腺癌患者。qRT-PCR检测化疗不同时间的血清miRNA表达,分析它们与化疗反应和预后的关系。结果患者血清miR-21、miR-125b、miR-155表达水平在化疗前显著高于健康女性(P <0. 01),miR-122与健康人无显著差别。化疗期间,血清miR-125b的表达和miR-21水平的改变与化疗反应和无瘤生存率(DFS)相关。化疗有效者miR-125b的表达在化疗前和化疗过程中均低于无效者(P <0. 01);化疗有效者miR-21的表达在化疗期间低于化疗前(P <0. 05)。miR-125b低表达的患者比高表达的患者有更好的DFS(P <0. 01); miR-21表达下降的患者比升高的患者有更好的DFS(P <0. 01)。结论血清miR-21和miR-125b能够作为乳腺癌新辅助化疗反应和预后的预测标志物。
【Abstract】 Objective To explore the predictive value of serum miRNAs for the neoadjuvant chemotherapy effect and prognosis in breast cancer. Methods A total of 102 patients with breast cancer who were receiving the neoadjuvant chemotherapy were included. The expression of serum miRNAs at different time of the chemotherapy was detected by qRT-PCR and its relationship with treatment response and prognosis was analyzed. Results The expression of serum miR-21,miR-125 b and miR-155 was significantly higher in breast cancer patients( P <0. 01),and miR-122 did not differ from the expression in healthy volunteers. The expression of serum miR-125 b and the changes of miR-21 levels during the treatment were related with treatment response and disease-free survival( DFS). The expression of serum miR-125 b in the responders before chemotherapy and during the process of it was significant lower than the non-responders( P < 0. 01). Serum miR-21 expression in the responders during the treatment was lower than before chemotherapy( P < 0. 05). Patients with lower expression of miR-125 b had better DFS than those with higher expression( P < 0. 01),and those with decreased serum miR-21 expression had favorable DFS than those with increased expression( P < 0. 01). Conclusion Serum miR-21 and miR-125 b are predictive markers for the neoadjuvant chemotherapy response and prognosis in breast cancer.
