麻芍平喘汤对哮喘模型大鼠气道重塑的干预作用
Intervention Effect of Mashao Pingchuan Decoction(麻芍平喘汤) on Airway Remodeling in Asthmatic Model Rats
【摘要】 目的:观察麻芍平喘汤对哮喘大鼠肺组织转化生长因子β1(TGF-β1)、α-平滑肌肌动蛋白(α-SMA蛋白)的影响,初步探讨麻芍平喘汤防治哮喘气道重塑的可能作用机制。方法:采用10%卵白蛋白加氢氧化铝干粉10 mg致敏和激发,建立哮喘大鼠气道重塑模型,将70只大鼠随机分为正常组、模型组、麻芍平喘低剂量组、麻芍平喘中剂量组、麻芍平喘高剂量组、地塞米松组、阳和平喘组,造模成功后,采用麻芍平喘汤、地塞米松和阳和平喘汤进行干预,干预结束后,HE染色法观察气道病理切片,肺功能测定仪测定用力呼气肺活量(FVC)、0.3 s用力呼气容积(FEV0.3)、最大呼气流量(PEF)与0.3 s用力呼气容积占用力呼气肺活量的比值(FEV0.3/FVC),RT-PCR法检测TGF-β1mRNA、α-SMA mRNA在肺组织中的表达。结果:模型组大鼠FVC、FEV0.3、PEF明显低于正常组(P<0.01),FEV0.3/FVC值明显高于正常组(P<0.01)。各给药组大鼠FVC、FEV0.3、PEF均有改善(P<0.05);模型组大鼠肺组织TGF-β1mRNA、α-SMA mRNA蛋白表达水平明显高于正常组,各给药组灰度值低于模型组(P<0.01),麻芍平喘高剂量组和阳和平喘组与地塞米松组比较,差异无统计学意义(P>0.05)。结论:麻芍平喘汤能够改善肺功能,降低TGF-β1、α-SMA的蛋白表达,抑制支气管平滑肌增生,减轻气道壁厚度,从而减轻气道炎症,延缓气道重塑。
【Abstract】 Objective: To observe the effect of Mashao Pingchuan Decoction on TGF-β1 and α-SMA protein in lung tissue of asthmatic rats, and to explore the possible mechanism of Mashao Pingchuan Decoction in preventing and treating airway remodeling of asthma. Methods: The airway remodeling model of asthmatic rats was established by sensitization and stimulation with 10% ovalbumin plus 10 mg aluminum hydroxide dry powder.Seventy rats were divided into seven groups namely normal group, model group, Masaopingchuan low dose group,Masaopingchuan medium dose group, Masaopingchuan high dose group, Dexamethasone group, Yanghe Pingchuan group. After successful modeling, Mashao Pingchuan Decoction, Dexamethasone and Yanghe Pingchuan Decoction(阳和平喘汤) were used for intervention. After the intervention, pathological sections of airway were observed by HE staining. The ratio of FVC, FEV0.3, PEF and FEV0.3/FVC was measured by pulmonary function analyzer. RTPCR was used to detect the expression of TGF-β1 mRNA and α-SMA mRNA in lung tissue. Results: FVC,FEV0.3 and PEF in the model group were significantly lower than those in the normal group(P <0.01), and FEV0.3/FVC was significantly higher than those in the normal group(P<0.01). FVC, FEV0.3 and PEF were im-proved in each group(P<0.05). The expression level of TGF-β1 mRNA and α-SMA mRNA in lung tissue of model group was significantly higher than that of normal group, and the gray value of each administration group was lower than that of model group(P<0.01). There was no significant difference between the high dose group and Yanghe Pingchuan Group and dexamethasone group(P>0.05). Conclusion: Mashao Pingchuan Decoction could improve lung function, decrease the protein expression of TGF-β1 and α-SMA, and inhibit the proliferation of bronchial smooth muscle and reduce the thickness of airway wall, thus reducing airway inflammation and delaying airway remodeling.
