白及疫病病原菌的生物学特性及侵染特性测定
Biological Characteristics and Infection Characteristics of the Pathogen of Bletilla striata Blight
【摘要】 为了明确危害白及的烟草疫霉与以往其他寄主上的烟草疫霉在生物学特性和侵染特性上的异同,试验以烟草疫霉病病原菌Bjphy4菌株为材料,进行了白及疫病病原的生物学特性和侵染特性研究。结果表明:该病菌的生长和产孢最适培养基为V8培养基,最适氮源为蛋白胨,菌丝生长和产孢子囊的最佳碳源不同,葡萄糖作碳源时最适宜菌丝生长,而淀粉作碳源时最利于病菌产孢子囊,最适pH值为7,最佳培养温度为25~30℃,最适光照条件为全黑暗培养;病菌菌丝致死温度为47℃;白及疫病病原的侵染特性测定结果显示,28℃为病菌的最佳侵染温度,相对湿度越高越利于病菌侵染,超过90%时适宜发病,全黑暗情况下更利于病菌侵染。
【Abstract】 In order to clarify the differences of biological and infection characteristics of Phytophthora nicotianae between Bletilla striata and on other hosts,the effects of culture medium,temperature,pH value,light,carbon and nitrogen sources on mycelium growth and sporulation of Phytophthora nicotianae were determined by using the pathogen strain Bjphy4 as material.The pathogen was inoculated on Bletilla striata at the stage of 4-6 leaves,the disease spot extension under different temperature,humidity and light conditions were observed.The results showed that:V8 was the optimal medium for mycelial growth and sporulation,and peptone was the most suitable nitrogen source.The optimum carbon sources for mycelial growth and sporulation were different.Glucose was beneficial to the mycelium growth,and starch promoted the sporangium formation.The other optimum conditions were pH 7,25~30℃ and full dark culture.The lethaltemperature of mycelia was 47℃.The optimal temperature for pathogen infection was 28℃.The infection was enhanced as relative humidityincreased,when humidity was>90%,it was suitable for the disease occurrence.Full dark condition was more favorable for the pathogen infection.
