响应曲面法优化氯乙酰儿茶酚的合成工艺

【摘要】 卡巴克络是一种常规止血药,治疗能力优越,故在中国乃至全球范围内市场需求量巨大。运用Design Expert软件中响应曲面法对氯乙酰儿茶酚的合成工艺条件进行优化分析,采用Box-Behnken设计实验优化原料物质的量比(邻苯二酚∶氯乙酰氯)、反应温度、反应时间对合成产率的影响,分析出影响因素的显著性和影响因素之间交互作用,优化出最佳的合成工艺条件,最后对产物进行表征。通过实验验证得出最优的工艺条件为:邻苯二酚与氯乙酰氯物质的量比为0.9,反应温度为44.05℃,反应时间为4.23 h,产物产率为75.606 8%。按照优化工艺进行合成的实际产物产率为75.03%,接近理论预测值。

【Abstract】 Carbazochrome is one of the conventional hemostatic drugs with superior therapeutic ability, so there is a huge market demand in China and even in the world. The RSM(response surface method)in Design Expert software was used to optimize the synthesis conditions of chloracetyl catechol. The Box-Behnken design experiment was used to optimize the effects of raw material molar ratio(catechol∶ chloroacetyl chloride), reaction temperature and reaction time on the synthesis yield, and the obvious influencing factors were analyzed. The best synthesis conditions were optimized by the interaction between the properties and influencing factors. Finally, the product was characterized. The optimal process conditions are as follows: the molar ratio of catechol to chloroacetyl chloride is 0.9, the reaction temperature is 44.05 ℃, the reaction time is4.23 h, and the product yield is 75.606 8%. According to the optimized process, the yield of the actual synthesized products is 75.03%, which is close to the predicted result.
