酸性手性药物在直链淀粉手性固定相上的拆分
Separations of Acid Chiral Drugs on Amylose Stationary Phase in HPLC
【摘要】 本文采用实验室自合成的直链淀粉三(3,5-二甲基苯基氨基甲酸酯)(ADMPC)涂覆的4种不同衍生化大孔硅胶以及纯大孔硅胶制成5种手性固定相。考察了4种流动相下5种手性柱对布洛芬、扁桃酸、DNB-亮氨酸、酮洛芬和氟比洛芬的拆分。这5种酸性手性药物在5种手性柱上都有不同程度的拆分,不同样品的最佳拆分条件有一定差异,ADMPC涂覆在反相大孔硅胶上对5种酸性手性药物的整体拆分效果优于氨基化大孔硅胶和纯大孔硅胶。为这五种手性药物的拆分提供了参考。
【Abstract】 In this paper, Amylose tri(3,5-dimethyl phenyl carbamate)(ADMPC) was synthesized and coated on pure or derivative macroporous silica gel to obtain five chiral stationary phases. With four mobile phases,ibuprofen, mandelic acid, DNB-leucine, ketoprofen and flurbiprofen was resolved on those chiral columns. The experimental results confirm that five chiral drugs have different degrees of resolution on five chiral columns, and the optimal separation conditions of different samples are different. ADMPC coated on macroporous silica gel usually has a good resolution. This research provides a way for the actual resolution of the five chiral drugs.
