白芨假鳞茎芽无菌苗高效增殖影响因子初步筛研究
Primary selections of factors to affect efficient sterile proliferation of pseudobulb Bletilla striata buds
【摘要】 以白芨(Bletilla striata)无菌苗材料为外植体,在不同培养基上,通过不同外源添加物、接种密度、活性炭浓度、切割手法对白芨假鳞茎芽无菌苗增殖影响试验,研究提高白芨假鳞茎芽无菌苗增殖系数的因子,以确定最适宜增殖条件.结果表明,在不同外源添加物的试验中,白芨假鳞茎芽无菌苗增殖的最适宜培养基为:MS+BA 2. 0 mg/L+NAA 0. 2mg/L+琼脂7. 0 g/L+香蕉.试验中以MS+BA 2. 0 mg/L+NAA 0. 2 mg/L+琼脂7. 0 g/L+蔗糖30 g/L为基础培养基,其中接种密度为6、活性炭浓度为3 g/L,交叉相切的切割手法,接种密度为4的同时中间横划一刀,均能显著提高白芨假鳞茎芽无菌苗的增殖系数,最高可以达到4,本实验为白芨快速繁殖和工厂化生产提供了技术支持.
【Abstract】 By using the tissue culture seedling of Bletilla striata as explant,culturing factors were investigated by adding different exogenous additives, regulating inoculum density and activated carbon concentration,employing different cutting technique and using different culturing media,to determine the optimum conditions for improving the proliferation coefficient of pseudobulb Bletilla striata buds. The results showed that in the experiments using different exogenous additives,the suitable medium for the proliferation of pseudobulbs buds was MS + BA 2. 0 mg/L + NAA 0. 2 mg/L + agar gel 7. 0 g/L + banana. In addition,MS +BA 2. 0 mg/L + NAA 0. 2 mg/L + agar gel 7. 0 g/L + sucrose 30 g/L used as the basic medium,combining the inoculation density 6,the concentration of activated carbon 3 g/L,and cross-cutting method, the inoculation density being 4 while the middle crossing a knife line,it can significantly improve the proliferation coefficient of the pseudobulb Bletilla striata buds,the maximum proliferation coefficient can reach to 4. This experiment provides technical support for rapid propagation and industrialization development of Bletilla striata.
