硫喷妥钠对心肌缺血-再灌注损伤大鼠心肌Toll样受体4和核因子-κB表达的影响

【摘要】 目的探讨硫喷妥钠对心肌缺血-再灌注损伤的大鼠心肌组织Toll样受体4(TLR4)/核因子-κB(NF-κB)信号通路的影响。方法 20只健康SD大鼠制备大鼠(I/R)动物模型随机分为假手术组(sham组)、缺血-再灌注组(I/R组)、缺血-再灌注+硫喷妥钠组(I/R+T组)、缺血-再灌注+硫喷妥钠组+TLR-4激活剂组(I/R+T+Neoseptin 3组),qRT-PCR、Western blot和ELISA实验分别检测4组大鼠心肌组织中TLR-4、NF-κB在mRNA和蛋白水平的表达及肿瘤坏死因子-α(TNF-α)和白介素1β(IL-1β)水平。结果与sham组比较,I/R组大鼠心肌组织中TLR-4、NF-κB在mRNA和蛋白水平的表达均显著上调,差异有统计学意义(P<0.05)。经I/R处理的I/R组大鼠心肌组织TNF-α和IL-1β水平显著高于sham组,差异有统计学意义(P<0.05)。I/R+T组大鼠心肌组织中TLR-4、NF-κB在mRNA和蛋白水平的表达显著低于I/R组,差异有统计学意义(P<0.05)。经硫喷妥钠麻醉后,I/R模型大鼠心肌组织TNF-α和IL-1β水平显著低于I/R组,差异有统计学意义(P<0.05)。I/R的大鼠心肌组织TLR-4、NF-κB蛋白水平,TNF-α和IL-1β水平显著高于sham组(P<0.05);而I/R+T组较I/R组大鼠心肌组织TLR-4、NF-κB表达水平显著下调(P<0.05),TNF-α和IL-1β水平显著降低(P<0.05);I/R+T+Neoseptin 3组大鼠心肌组织中TLR-4、NF-κB表达及TNF-α和IL-1β水平基本恢复至I/R组水平(P<0.05)。结论硫喷妥钠能够通过降低心肌缺血-再灌注损伤大鼠心肌组织中TLR-4和NF-κB表达,抑制炎性反应,从而保护心肌,减轻心肌-缺血再灌注损伤。

【Abstract】 Objective To investigate the effects of thiopental sodium on the expression levels of Toll-like receptor 4 and nuclear factor-κB in rats with myocardial ischemia-reperfusion injury.Methods Twenty normal SD rats were randomly divided into four groups:sham operation group, ischemia reperfusion injury group(I/R group), I/R+thiopental sodium group(I/R+T group) and I/R+T+Neoseptin 3 group.The qRT PCR,Western blot and ELISA were used to detect the expression levels of TLR4 and NF-κB mRNA and protein in myocardium tissues of rats,and the levels of TNF-α and IL-1β, respectively.Results As compared with those in sham group, the expression levels of TLR-4 and NF-κB in myocardium tissues of rats in I/R group were significantly increased(P<0.05). The levels of TNF-α and IL-1β of myocardium of rats in I/R group after management of ischemia reperfusion were significantly higher than those in sham group(P<0.05). Moreover, the levels of TLR4 and NF-κB mRNA and protein in myocardium of rats in I/R+T group were significantly lower than those in I/R group(P<0.05). After anesthesia by using thiopental sodium, the levels of TNF-α and IL-1β of myocardium in I/R model rats were significantly lower than those in I/R group(P<0.05). The protein levels of TLR-4 and NF-κB as well as the levels of TNF-α and IL-1β in myocardium of I/R model rats were significantly higher than those in sham group(P<0.05). Furthermore,as compared with those in I/R group, the expression levels of TLR-4, NF-κB and the levels of TNF-α and IL-1β in I/R+T group were significantly decreased(P<0.05). In addition,the expression levels of TLR-4 and NF-κB as well as the levels of TNF-α and IL-1β in I/R+T+Neoseptin 3 group restored to the levels in I/R group.Conclusion Thiopental sodium can protect myocardium and reduce myocardial ischemia-reperfusion injury by inhibiting inflammation via decreasing the expression levels of TLR-4 and NF-κB,so as to protect myocardium and relieve the myocardial ischemia-reperfusion injury in rats.
