USP1调节心肌细胞肥大的效果研究
Experimental study on the effects of USP1 in regulating myocardial hypertrophy
【摘要】 目的研究泛素特异性蛋白酶1(USP1)在心肌肥厚中的作用。方法建立异丙肾上腺素(ISO)诱导的小鼠心肌肥厚模型并应用荧光定量PCR方法及Western Blot方法检测USP1在正常小鼠心脏组织及心肌肥厚小鼠心脏组织中的表达情况。分离大鼠原代心肌细胞,并用腺病毒的方法干扰USP1的表达,应用荧光定量PCR方法及Western Blot方法检测USP1的干扰效率;应用Image J软件统计干扰USP1后心肌细胞的面积,并应用荧光定量PCR方法检测下调USP1的表达对心肌肥厚相关基因Anp及Bnp mRNA表达的影响。结果 ISO诱导的小鼠心脏组织中心肌肥厚标志性基因Anp、 Bnp及Myh7的mRNA表达水平显著上调,提示心肌肥厚小鼠模型建立成功;USP1在小鼠心肌肥厚的心脏组织中其蛋白水平及mRNA水平均显著上调(P<0.01);荧光定量PCR及Western Blot结果显示,在大鼠原代心肌细胞中USP1得到有效敲低;干扰USP1的表达后,原代心肌细胞面积减小,并且抑制心肌肥厚标志物Anp及Bnp基因的表达。结论干扰USP1可以抑制心肌肥厚的发生。
【Abstract】 Objective To investigate the effects of ubiquitin-specific peptidase 1(USP1) in mice with myocardial hypertrophy.Methods The mice models with isoproterenol(ISO)-induced myocardial hypertrophy were established,and the expression levels of USP1 in normal mice heart tissues and the myocardial tiisues of mice with myocardial hypertrophy were detected by fluorescence quantitative PCR and Western Blot.Primary myocardial cells of rats were isolated, and the expression of USP1 was interfered by adenovirus.The interference efficiency of USP1 was detected by fluorescence quantitative PCR and Western Blot.Image J software was used to count the area of myocardial cells after knockdown of USP1,and fluorescence quantitative PCR was used to detect the effects of USP1 knockdown on the mRNA expression of myocardial hypertrophy marker genes-Anp,Bnp and Myh7.Results The expression levels of Anp mRNA,Bnp mRNA and Myh7 mRNA in ISO-induced mice heart tissues were significantly up-regulated,which suggested that the animal model with myocardial hypertrophy were established successfully. The protein levels and mRNA levels of USP1 were significantly up-regulated in cardiac tissues of mice with myocardial hypertrophy.The fluorescence quantitative PCR and Western Blot results showed that USP1 was effectively knocked down in primary myocardial cells of rats,and the USP1 knockdown decreased the cell surface area of primary myocardial cells and significantly inhibited the mRNA expression of Anp gene and Bnp gene.Conclusion The interference of USP1 can inhibit the pathogenesis of myocardial hypertrophy in mice.
