几种化学因素对海蜇毒素溶血活性的稳定效果
The stabilizing effect of several chemical factors on the hemolytic activity of jellyfish toxin
【摘要】 为解决在放置一段时间后海蜇(Rhopilema esculentum)毒素溶血活性锐减的问题,利用分光光度计测量了EDTA、GSH、Ca2+、NaCl等化学因素对海蜇毒素溶血活性的稳定效果。结果发现:(1)EDTA对毒素具有较好的稳定作用,浓度在0.5 mmol/L时溶血活性最高,其溶血率为93.63%;(2)NaCl对毒素也具有稳定作用,当浓度在5 mmol/L时溶血活性最高,其溶血率为92.68%;(3)GSH对毒素稳定作用较差,在浓度为0.5 mmol/L时溶血活性最高,其溶血率为58.17%;(4)Ca2+对海蜇毒素不具有稳定作用;(5)正交试验发现,NaCl对溶血活性的稳定性影响最大,在GSH、NaCl、EDTA浓度分别为1.0、5、0 mmol/L时,海蜇毒素的溶血性最强。
【Abstract】 In order to solve the problem that the hemolytic activity of jellyfish(Rhopilema esculentum) toxin decreases sharply after being placed for a period of time, the stabilizing effect of chemical factors such as EDTA, GSH, Ca2+ and NaCl on the hemolytic activity of jellyfish toxin was measured by spectrophotometer. The results indicated that:(1) The presence of EDTA benefited the stability of the hemolytic activity. The hemolytic activity was the highest when the concentration was 0.5 mmol/L, and its hemolysis rate was 93.63%.(2) NaCl also had a stabilizing effect on the toxin. When the concentration was 5 mmol/L, the hemolytic activity was the highest and the hemolysis rate was 92.68%.(3) GSH had a poor stabilizing effect on the toxin. The hemolytic activity was the highest when the concentration was 0.5 mmol/L, and its hemolysis rate was 58.17%.(4) The results showed that Ca2+ had no stabilizing effect on jellyfish toxin.(5) Orthogonal experiment found that NaCl had the greatest influence on the stability of hemolytic activity. When the concentrations of GSH, NaCl, and EDTA were 1.0, 5, and 0 mmol/L,respectively, the hemolysis of jellyfish toxin is the strongest.
